Do I have to wait for EZ2 Connect Fx to cool down before starting a new run?
No, one doesn’t have to wait for the cooldown of the device for a re-run with Sep&Prep. When starting a run to process non-sperm or any other samples with the EZ1&2 DNA Investigator Kit shortly after an Investigator Sep&Prep run, the heating block of the instrument is still at high temperature. This may cause increased evaporation of elution buffer, which is transferred to the heating position after a protocol is started, resulting in slightly lower eluate volumes. If this is observed, wait until the heating block has cooled to a lower temperature. Note that the elution buffer is heated to 65°C during a run. Thus, cooling to lower temperatures is not required.

Sexual assault samples are commonly comprised of a mixture of non-sperm and sperm cells from multiple individuals. For purification of the non-sperm fractions (NSF), we recommend the use of EZ1&2 DNA Investigator Kit which can either be done directly afterwards on the same EZ2 Connect Fx instrument or at a later timepoint.

The EZ1&2 DNA Investigator Kit reproducibly automates purification of genomic DNA from reference and casework samples in human identity testing. The kit can be used with the EZ2 Connect Fx (1-24 samples), EZ1 Advanced XL (1–14 samples per run), EZ1 Advanced (1–6 samples per run), and BioRobot EZ1 (1–6 samples per run) instruments. Purification is fast and efficient. Purified DNA performs well in downstream analyses with high signal-to-noise ratios, such as quantitative PCR and STR analysis.
