Performance

MinElute 96 UF PCR Purification Kit使用96孔板高通量纯化PCR产物。在BioRobot 3000(已停产)、BioRobot 8000或BioRobot Universal System上全自动处理1–8个96孔板,或在QIAvac Multiwell Manifold(货号:9014579,需合适的真空管,如QIAvac Multiwell管,建议使用孔板振荡器)上处理1个96孔板。纯化>100 bp的DNA的回收率为80–95%。使用少量洗脱液(人工操作需20 µl,如使用BioRobot工作站则需30 µl)即可将纯化的PCR产物从膜表面洗脱下来,得到高浓度DNA(参见 Highly concentrated DNA eluates)用于各种下游应用。无论是人工操作还是全自动流程都可处理多达150 µl的PCR产物。

Principle

QIAGEN MinElute 96 UF PCR Purification Kit设计独特,结合了多孔技术的便利性和超滤膜的高效分离特性。使用孔板可对>100 bp的双链DNA片段进行快速高效回收,并除去引物(<20mers)、未整合的核苷酸链和盐。

Procedure

MinElute 96 UF PCR Purification Kits运用超滤技术,在真空环境下使用QIAvac纯化PCR产物(参见" MinElute 96 UF PCR Purification procedure")。在真空条件下,将PCR产物上样超滤板的孔中。引物、盐和未整合的核苷酸链等小分子通过滤膜,而大于100 bp的PCR产物留下来。水、DMSO或缓冲液如SSC等可以作为洗脱液。MinElute 96 UF PCR Purification操作流程可在BioRobot 3000(已停产)、BioRobot 8000、或BioRobot Universal System上全自动进行。

Can MinElute 96 UF PCR be used to purify PCR samples which contain mineral oil?
We do not recommend the use of samples containing mineral oil since mineral oil will not be removed during the purification procedure.

Can I use a partial MinElute 96 UF plate?

Yes, you may use part of the MinElute 96 UF Plate, and use the rest of the plate later. In contrast to silica membrane based plates, the unused part of the UF plate does not need to be taped off. We do not recommend to re-use MinElute 96 UF Plates more than 2-4 times.
