QIAsymphony RNA Kit は、QIAsymphony SP上で1～96 サンプルからのトータルRNAあるいはmiRNAやその他のsmall RNAを含むトータルRNAの完全自動化精製を実現します。微量のスタートサンプル（細胞100 個以上）から最高1 x 10⁷個の細胞あるいは50 mg の組織を用いて動物細胞、組織、FFPE組織からトータルRNA を精製可能です。至適化済みのプロトコールにより高純度のRNAまたはmiRNAを含むRNAを高収量で得られ、溶出容量が調節可能なので各ダウンストリームに応じて容易にRNA 濃度を至適化できます。充填済み試薬によりセットアップ時間を短縮でき、マニュアルでの作業操作も最小限に抑えることができます。バーコードの読み取りにより試薬の使用履歴のトラッキングが可能です。

QIAsymphony SP上でのRNA自動化精製では、培養細胞、組織、FFPE組織から高い再現性で質の高いRNAを高収量で得ることができ、マニュアル精製操作で得られるRNAと同等のRNAを回収できます（図 ""）。RNA精製用のQIAsymphony protocol では、主としてmRNAを含む200ヌクレオチドより長いRNA分子が多く含まれています（図 ""）。QIAsymphony miRNA protocol を使用すれば、miRNAsや他の低分子RNAなどの200ヌクレオチドより短いRNAを含むトータルRNAを精製できます（図 ""）。

Principle

QIAsymphony RNA Kitは、proteinase K処理後に200ヌクレオチドより長いトータルRNAやmiRNAおよびその他の低分子RNAを含むトータルRNAを精製するために、シリカベーステクノロジーのスピードと効率と磁性粒子による簡便な操作を組み合わせています（図 ""）。スタートサンプルに、動物の細胞や組織も使用できます。追加の製品を使えば、ホルマリン固定パラフィン包埋（FFPE）組織も本キットで処理できます。サンプルを溶解し、カオトロピック塩の存在下で粒子のシリカ表面へ結合させることによりライセートからRNAを1ステップで精製します。マグネットを使用してライセートから粒子を分離させ、DNAはRNaseフリーのDNase処理により取り除きます。磁性粒子を効率的に洗浄し、RNAをRNaseフリー水で溶出します。

RNA was purified from Jurkat cells on the QIAsymphony SP using the QIAsymphony RNA Kit. RNA yields from 2 instrument runs using 2 different reagent cartridge lots were compared with manual purification using the RNeasy Mini Kit. Each bar represents the average of 4 samples.

RNA processed on the QIAsymphony SP with the QIAsymphony RNA Kit can be eluted in 200, 100, or 50 µl.

Elution in smaller volumes will provide higher concentration, but lower overall yield. Total RNA yield with elution in 50 µl will be about 20% lower compared to elution in 100 µl.

Yes, the QIAsymphony RNA Kit is compatible with stabilized samples. We recommend stabilizing tissue samples with RNAprotect Tissue Reagent or AllProtect Tissue Reagent, or by flash-freezing. Cultured cells can be lysed directly in cell culture dishes, or after pelleting. Alternatively, cells can be stabilized using RNAprotect Cell Reagent.

The QIAsymphony RNA Kit enables purification of RNA from cultured cells and easy-to-lyse tissues, such as kidney, liver, and spleen on the QIAsymphony SP. A large-volume protocol is available that enables processing of larger sample amounts.

Cultured cells: up to 3x10⁶ cells (standard protocol); up to 1x10⁷ (large volume protocol)
Tissues: up to 20 mg of most tissues (standard protocol); up to 50 mg (large-volume protocol)

For particularly DNA-rich tissues, such as spleen or thymus, we recommend either using no more than half the maximum amount, or using DNase at a higher concentration (see the QIAsymphony RNA Handbook; additional DNase required).

Strongly denaturing conditions in the lysis buffer (Buffer RLT Plus) of the QIAsymphony RNA Kit keep RNases in an inactive state. Keeping homogenates on the QIAsymphony instrument at room temperature (20-25°C) for over 6 hours does not impair RNA integrity.

If only the large-volume protocol is used, the number of samples that can be prepared per kit is reduced to 96 (48 per reagent cartridge). Any combination of standard and large-volume sample preparation can be performed.
