Product Details

DNeasy mericon Food Kit 设计用于从各种生食品和加工食品中提取高品质的核酸。可尽量减少从复杂的食品基质中携带 PCR 抑制物的情况。该试剂盒是综合性 QIAGEN 食品检测产品组合的一部分,后者还具有针对病原体和 GMO DNA 检测以及成分认证的检测。操作流程可在 QIAcube Connect 上全自动完成。

Performance

在生产过程中,食品会受到高温、辐照、高压和 pH 值变化的影响,从而导致 DNA 降解和断裂。DNeasy mericon Food Kit 的化学性质经过优化,可回收短 DNA 片段(最小 100 bp),确保即使是高度片段化的 DNA 也能得到高效提取,并随后在 PCR 反应中进行扩增。凭借这些特点,使用 DNeasy mericon Food Kit 进行提取成为第一种普遍适用的提取方法,即使在使用强抑制性、高度加工、多脂肪、酸性、高或低 DNA 含量的食品时,也能获得非常不错且可靠的结果(见图“DNA 产量高,携带抑制物极少”和图“对抑制物的耐受性提高”)。此外,不再需要针对不同的食品基质制定多种裂解操作流程。

Principle

DNeasy mericon Food Kit 采用改良的十六烷基三甲基溴化铵 (CTAB) 提取细胞总核酸。

非离子洗涤剂 CTAB 广泛用于从多种组织类型中高效提取细胞总核酸。根据盐分条件的不同,CTAB 可与细胞核酸(低盐条件)或多糖、蛋白质和植物代谢物等细胞抑制物(高盐条件;如 Food Lysis Buffer 中的情况)复合。

与传统 CTAB 方案相比,其工作流程步骤更少,2.5 小时内可处理多达 30 个样本(见图“高效快速的 DNA 提取”)。使用这些试剂盒进行 DNA 回收的回收率和效率明显更高。

Procedure

DNeasy mericon Food Kit 的优化方案将 CTAB 与 Proteinase K 结合使用,首先消化致密组织,然后沉淀蛋白质,同时沉淀其他细胞和食源性抑制物。

离心沉淀抑制物,提取的 DNA 仍留在溶液中。在随后的氯仿萃取过程中,未沉淀的任何残留 CTAB 蛋白、CTAB 碎片或 CTAB 多糖复合物与其他亲脂性抑制物一起通过萃取到有机氯仿相中而被去除。只有含有 DNA 和已明显耗尽抑制物的水相才会得到进一步处理。将该水相与结合缓冲液(用于调整结合条件)混合后应用于 QIAquick Spin Columns。得到的 DNA 可立即用于下游 mericon real-time PCR 检测。

Why is my 260/280 ratio low after using the DNeasy mericon Food Kit?
With complex food samples, a classical evaluation of DNA purity and protein content by A260/A280 ratio is not a significant criterion. Due to inevitable traces of food matrix in the sample background, a spectroscopic evaluation of DNA or protein absorptions is highly unreliable, and we have found that there is not a correlation between 260/280 and 260/230 ratios and PCR performance. Even DNA with Low 260/280 and 260/230 can produce good PCR results.

Does salt concentration in the food matter when using the DNeasy mericon Food Kit?
We have not observed any effect of NaCl concentration in the food on the effectiveness of the DNeasy mericon Food Kit.

CAn I use milk or other liquids with the DNeasy mericon Food kit? What volume would I use?
Liquids can be used. Use 2 ml (for 2 g protocol) or 200 ul (for 200 mg protocol) and mix it with the Food Lysis Buffer and Proteinase K solution as in the normal protocol.

Why does the DNeasy mericon Food Kit use a QIAquick column and Buffer PB rather than a DNeasy column and Buffer AL, which might be expected since the kit isolates genomic DNA?
During kit development, we determined that with the chemistry used in the kit and the tested sample material, the QIAquick column together with the buffer PB had the best performance.
