Performance

Principle

QIAamp DNA Investigator Kit 将硅胶膜的选择性附着属性与 20 至 100 µl 的灵活洗脱体积相结合。

Procedure

QIAamp DNA Investigator 使用 QIAamp MinElute Spin Columns 纯化高质量 DNA,洗脱体积灵活,工作流程包括 4 个步骤(见“ 程序”流程图)。样本在变性条件下用蛋白酶 K 裂解。DNA 与膜结合后,污染物流过,残留污染物被洗去。浓缩的纯 DNA 从膜上洗脱下来。QIAamp DNA Investigator Kit 可在  QIAcube 上自动运行。

Short-term storage (up to 4 weeks) at room temperature (15–25°C) does not affect the performance. However, for optimal performance and quality, storage temperature should not exceed 25°C.

What can be used as an alternative to the A260 measurement for quantification of small amounts of RNA and DNA?

Small amounts of RNA and DNA may be difficult to measure spectrophotometrically. Fluorometric measurements, or quantitative RT-PCR and PCR are more sensitive and accurate methods to quantify low amounts of RNA or DNA.

Fluorometric measurements are carried out using nucleic acid binding dyes, such as RiboGreen® RNA Quantitation Reagent for RNA, and PicoGreen® DNA Quantitation Reagent for DNA (Molecular Probes, Inc.).
