MiSeq FGx Sequencing Reagents support both Forensic Genomics and Research Use Only modes on the MiSeq FGx Sequencing System to simplify inventory and ordering for all applications.

Performance

The MiSeq FGx Reagent Kit has been optimized for sequencing forensic samples and delivers high-quality performance for a range of forensic applications. The extended read lengths enable deep sequencing of STRs, mtDNA and SNPs. The MiSeq FGx Reagent Micro Kit is purpose-built for lower throughput and faster sequencing time, while delivering equivalent quality. Both kit sizes are quality controlled to meet the high standards expected from forensic laboratories. The MiSeq FGx Sequencing Reagents are integral to the MiSeq FGx Sequencing System and support analysis of libraries across the range of Verogen NGS kits.

Principle

The MiSeq FGx System achieves exceptional data quality by employing a proprietary, reversible terminator‑based method that detects single bases as they are incorporated into massively parallel DNA strands. Fluorescent terminator dyes are imaged as each deoxynucleotide triphosphate (dNTP) is added and then cleaved to allow incorporation of the next base. With all four reversible, terminator-bound dNTPs present at each sequencing cycle, natural competition among bases minimizes incorporation bias.

The MiSeq FGx Reagent Micro Kit and the MiSeq FGx Reagent Kit are identical in manufacture. With both kits, the best-in-class sequencing-by-synthesis (SBS) technology remains unaltered. The only difference is the surface area of the flow cell available for imaging. The MiSeq FGx System images the micro flow cell at approximately 35% of the standard flow cell area for a total output of 5 million reads.

Procedure

The first sequencing step of the MiSeq FGx Sequencing System uses a library prep kit to add primers to genomic DNA (gDNA), mtDNA, or mRNA extracted from forensic samples for simultaneous capture and amplification of hundreds of target regions, generating dual-indexed libraries ready for sequencing. After library prep, load-and-go MiSeq FGx Sequencing Reagents streamline run setup. Simply thaw the prefilled reagent cartridge, add libraries to the cartridge, and insert the cartridge into the instrument. The system software accepts run parameters and starts sequencing with the push of a button.
