Product Details

QIAsymphony DSP DNA Kits, in combination with the QIAsymphony SP, enable automated total DNA purification from human whole blood, buffy coat, tissues and FFPE tissues, as well as viral DNA from human whole blood for in vitro diagnostic use. Kits are available in mini and midi formats for sample volumes ranging from 200 µl to 1000 µl. For tissue and FFPE tissue protocols, manual sample pretreatment is required. Optimized protocols provide high yields of pure DNA, and flexible elution volumes allow DNA concentration to be easily optimized for each downstream application. Novel, prefilled reagent cartridges reduce setup time and minimize manual handling. Bar code reading enables full tracking of reagents.

Principle

QIAsymphony DSP DNA Kits can be used to purify total DNA from human whole blood, buffy coat, tissues and FFPE tissues, as well as viral DNA from human whole blood.

QIAsymphony DSP DNA Kits combine the speed and efficiency of silica-based purification of DNA with the convenient handling of magnetic particles (see figure " Schematic of the QIAsymphony SP principle"). Innovative, ready-to-run reagent cartridges are prefilled with all reagents required for the purification procedure and are automatically opened by the QIAsymphony SP, which helps to minimize the risk of environmental contamination (see figure " Prefilled, sealed reagent cartridges"). Worktable setup is rapid, which saves you valuable time. Simply place up to 2 reagent cartridges in the consumables drawer. The reagent cartridges can be either from the same kit or from different kits, enabling different purification procedures to be performed within the same run of 96 samples. Reagent volumes only for the selected number of samples are used, giving you complete cost control.

Procedure

The purification procedure is designed to ensure safe and reproducible handling of potentially infectious samples, and comprises 4 steps: lyse, bind, wash, and elute (see figure "QIAsymphony DSP DNA procedure"). Optimized protocols provide high quality nucleic acids with efficient removal of proteins, nucleases, and other impurities. Nucleic acids purified with QIAsymphony DSP DNA Kits are ready for direct use in downstream applications such as amplification. The user can choose between different elution volumes.

FAQ

What sample types can be processed using the tissue protocols and the QIAsymphony DSP DNA Mini Kit?
Tissue and FFPE tissue can be processed using the tissue protocols and the QIAsymphony DSP DNA Mini Kit on the QIAsymphony SP.
FAQ ID -2925
How much starting material can be used with the QIAsymphony DSP DNA Mini Kit on the QIAsymphony SP?

Tissue: If no information about the expected yield is available, we recommend starting with 25 mg sample material. Depending on the yield obtained, the sample size can be increased in subsequent preparations to a maximum of 50 mg.

FFPE: Up to 4 sections, each with a thickness of up to 10 µm, or 8 sections with a thickness of up to 5 µm and a surface area of up to 250 mm2, can be combined in one preparation.

FAQ ID -2927
Do tissue samples require pretreatment before loading onto the QIAsymphony SP for DNA extraction?
Yes. Samples have to be completely lysed and homogenized prior to automated processing on the QIAsymphony SP platform using the QIAsymphony DSP DNA Mini Kit. Tissue samples are lysed by proteinase K digestion. FFPE tissue samples are deparaffinized with Deparaffinization Solution or xylene/ethanol and lysed by proteinase K digestion. Insoluble material is pelleted by centrifugation. Lysates are placed on the QIAsymphony SP for purification of DNA (see protocol sheets for details).
FAQ ID -2926
What sample volumes can be used with the tissue HC and LC protocols on the QIAsymphony SP?
After pretreatment, 220 µl of the lysates must be transferred to the QIAsymphony SP to ensure that 200 µl of sample is processed.
FAQ ID -2992
What are the recommended lysis times for tissue samples on the QIAsymphony SP?
Lysis time varies depending on the tissue type processed. For recommended lysis times refer to the tissue protocol sheets. If lysis is incomplete after recommended lysis time, as indicated by the presence of insoluble material or highly viscous lysates, lysis time can be prolonged or insoluble material can be removed by centrifugation. Overnight lysis is possible and does not affect the preparation.
FAQ ID -2993
What sample types should be processed with the low content (LC) and high content (HC) protocols for DNA extraction on the QIAsymphony SP?
Tissues will give increased DNA yields when processed with the high content protocol, but the low content protocol in combination with a small elution volume (50µl) may be used if high DNA concentration is required. For FFPE tissue we recommend using the low content protocol.
FAQ ID -2989
Will mitochondrial DNA also be isolated using the DNA tissue protocols and QIAsymphony DSP DNA Mini Kit on the QIAsymphony SP?
Yes. The DNA tissue protocols on the QIAsymphony SP purify genomic and mitochondrial DNA.
