Sensitive PCR using DNA purified from preserved blood.
PCR of a 900 bp fragment of the MECL-1 single-copy gene using serial dilutions of template DNA purified from 200 µl ACD-, heparin-, and EDTA-preserved whole blood. Purified DNA (15-0.25 µl) was used in each 50 µl PCR as indicated. M: 100 bp DNA ladder; C: negative control.
Reproducible performance in sensitive PCR analysis.
Amplification of a single-copy gene (MECL-1) using template DNA purified from whole blood. Upper and lower lanes are from PCR using template DNA from 200 µl and 350 µl blood samples, respectively. Lanes 1–6 and 7–12 are from the first and last of 8 processing runs on the BioRobot EZ1, respectively. Blood from the same donor is used in all isolations. M: 100 bp DNA ladder (100 ng); +: positive control; –: negative control. 2 µl (1%) of each PCR was loaded on the agarose gel.
