QIAsymphony DSP AXpH DNA Kit

For automated isolation and purification of DNA from liquid-based cytology media

Features

  • Novel, prefilled reagent cartridges for ease of use
  • High reproducibility through standardized processing
  • Continuous loading of up to 96 samples per run
  • Bar codes enable tracking of samples and reagents

Product Details

The QIAsymphony DSP AXpH DNA Kit, in combination with the QIAsymphony SP, enables automated DNA purification from liquid-based cytology media for in vitro diagnostic use. Optimized protocols provide DNA eluates that are ready to use in downstream applications. Novel, prefilled reagent cartridges reduce setup time and minimize manual handling. Bar code reading enables full tracking of reagents. The QIAsymphony SP performs all steps of the sample preparation procedure. Up to 96 samples, in batches of up to 24, are processed in a single run.

Principle

QIAsymphony DSP AXpH technology combines the speed and efficiency of pH-driven anion exchange chromatography with the convenient handling of magnetic particles (see " Schematic of the QIAsymphony SP principle"). Innovative, ready-to-run reagent cartridges are prefilled with all reagents required for the purification procedure and are automatically opened by the QIAsymphony SP, which helps to minimize the risk of environmental contamination.

Procedure

The purification procedure is designed to ensure safe and reproducible handling of potentially infectious samples, and comprises 4 steps: lyse, bind, wash, and elute (see " QIAsymphony DSP AXpH procedure"). Worktable setup is rapid, which saves valuable time. Simply place up to 2 reagent cartridges in the consumables drawer. The reagent cartridges can be either from the same kit or from different kits, enabling different purification procedures to be performed within the same run of 96 samples. Reagent volumes only for the selected number of samples are used, allowing complete cost control.

Applications

The QIAsymphony DSP AXpH DNA Kit in combination with the QIAsymphony SP enables purification of DNA from liquid-based cytology samples, such as cervical swabs collected in SurePath (BD) or PreservCyt (Cytyc/Hologic) media.

FAQ

How much starting material can be used with the QIAsymphony DSP DNA Mini Kit on the QIAsymphony SP?

Tissue: If no information about the expected yield is available, we recommend starting with 25 mg sample material. Depending on the yield obtained, the sample size can be increased in subsequent preparations to a maximum of 50 mg.

FFPE: Up to 4 sections, each with a thickness of up to 10 µm, or 8 sections with a thickness of up to 5 µm and a surface area of up to 250 mm2, can be combined in one preparation.

FAQ ID -2927
What is the input sample volume when using the QIAsymphony DSP AXpH kit?
The minimum input sample volume when using the QIAsymphony DSP AXpH kit is 2.5mL.   However, we recommend the use of a 4mL sample volume.
FAQ ID -2272
What downstream applications can I use the eluates from the QIAsymphony DSP AXpH DNA Kit?
The QIAsymphony DSP AXpH DNA Kit provides DNA eluates that are ready for direct use in downstream applications, such as hybridization-based assays or enzymatic reactions.  However, the eluates should not be used in PCR.
FAQ ID -2275
How long can the extracted DNA from the QIAsymphony DSP AXpH DNA Kit be stored?
For short-term storage of up to 24 hours, we recommend storing purified DNA in microtiter plates at 2–8ºC. For long-term storage of over 24 hours, we recommend storing purified DNA in elution microtube racks at –20ºC.
FAQ ID -2274
What is the intended use of the QIAsymphony DSP AXpH kit?
The QIAsymphony DSP AXpH DNA Kit utilizes AXpH technology for automated isolation and purification of DNA from biological specimens.
FAQ ID -2273
What is the throughput of the QIAsymphony when using the DSP AXpH protocol?
When using the QIAsymphony DSP AXpH kit, the QIAsymphony can process approximately 160 samples in an 8 hour shift.
