Product Details

dNTP Set consists of individual, highly pure deoxynucleotide triphosphates at 100 mM concentration in aqueous solution. Supply of each dNTP at high concentration and purity enables customized reaction setup for standard, sensitive and specialized PCR applications. High-purity dNTPs are essential for sensitive PCR where contaminating impurities may reduce sensitivity, amplification efficiency and product yield.

A set of individual dNTPs allows adjustments to the concentrations of specific nucleotides. This is necessary for certain labeling techniques, and desirable for flexibility to troubleshoot and alter reaction components.

A combined dNTP ready-to-use product is available. dNTP Mix contains dATP, dCTP, dGTP and dTTP in water (pH 7.5), each at a concentration of 10 mM. This product is suitable for use in all standard PCR techniques and sensitive PCR techniques.

Performance

The performance of each lot of dNTP Set is tested in a specific polymerase chain reaction requiring all four nucleotides.

Principle

dNTP Set is ideal for customized labeling and specialized PCR applications by allowing adjustment of individual deoxynucleotide ratios and concentrations.

Procedure

Prepare standard PCR assays with a typical 200 µM concentration of each dNTP. Manually formulate a specific dNTP mix to free up space for a labeled nucleotide in applications like custom labeling in standard PCR and RT-PCR using, for example, HotStarTaq or QIAGEN OneStep RT-PCR Kit, or in enzymatic labeling procedures with Klenow Fragment (3'→5' exo-). Minimize PCR carryover contamination by substituting dTTP with dUTP in the primary amplification and treating with thermolabile Uracil DNA Glycosylase (UDG) before the new PCR reaction starts.
