Adapt/develop a protocol

Before beginning troubleshooting or protocol development, check to see if a QIAGEN-validated QIAsprint Connect protocol already exists for your sample type.

If a protocol already exists, start with the QIAGEN-validated workflow and only adjust parameters if absolutely necessary. If no protocol exists, we recommend first characterizing your sample to determine which combination of modular QIAsprint Kits provides the best starting point.

Then, establish a manual proof-of-principle workflow to evaluate whether the selected PrepSet and Essential Kit combination is compatible with your sample. Once you have established a reliable manual workflow, the proven QIAGEN chemistry can be transferred to the QIAsprint Connect instrument with minimal effort.

Starting with manual testing is often the fastest and most efficient way to establish a new workflow. Manual purification allows rapid adjustment of conditions without the additional complexity of automation. It also enables direct observation of how the sample behaves during lysis and purification, which is critical when dealing with unusual viscosity or difficult-to-lyse material.

Importantly, manual testing makes it much easier to distinguish whether an issue is caused by the chemistry itself (e.g., lysis, binding or washing conditions) or by the automated processing. It also enables different parameters to be tested in parallel.

For magnetic bead-based methods, use magnetic stands that match your tube format to streamline bead separation and minimize hands-on time. As most manual method development work is performed in 1.5–2 mL tubes, the AdnaMag-S (for 1.5 mL tubes) or the MagAttract Magnetic Rack (for 2 mL tubes) are ideal. The racks support up to 12 samples in parallel and reliably immobilizes magnetic beads in 2 mL tubes. Comparable magnetic stands will work as well.

Once reliable yield and purity have been achieved manually with a defined combination of PrepSet and Essential Kit, the proven QIAGEN chemistry can be transferred to QIAsprint Connect with less effort. This approach significantly reduces trial-and-error testing on the instrument and accelerates the development of a robust automated protocol.
