What is the optimal DNA input and cycling conditions with the Investigator IDplex kit?

The optimal DNA input range is 200–500 pg, although reliable results can be obtained with <100 pg. If using higher amounts of input DNA, consider adjusting the number of PCR cycles.

  • 30 cycles (5+25) is standard. 32 cycles can be used for critical stains <100 pg (5+27), and 28 cycles for high DNA amounts >1 ng (5+23).

Up to 16.9 µl can be added to the reaction, provided that the water volume is adjusted accordingly.

  • Note that greater volumes of input DNA may allow potential PCR inhibitors to affect the PCR.
