Product Details

The MagAttract 96 DNA Plant Core Kit combines the efficiency of silica-based DNA purification with the convenient handling offered by magnetic particles, in manual, semi-automated, or automated 96-well plate format.

Purification using the MagAttract 96 DNA Plant Core Kit
Format:96-well sample plates
Sample sources:Plant tissue
Plant starting material, fresh or frozen:10–100 mg
Plant starting material, dried or lyophilized:10–30 mg
Typical yield:1–15 µg
Elution volume:100 µl

Purified DNA has been used successfully in PCR and real-time PCR (see figures " Successful PCR analysis" and " Reliable real-time PCR analysis").

See figures

The MagAttract 96 DNA Plant Kit is designed for fully automated, high-throughput preparation of genomic and chloroplast DNA from plant tissue. Proven MagAttract magnetic particle technology in combination with the BioRobot Plant Science system (no longer available) provides efficient and reproducible DNA purification (see figure " Reproducible yields").
See figures

Plant samples are mechanically disrupted using a homogenizer such as the TissueLyser. The resulting powder is resuspended in lysis buffer, thoroughly mixed, and centrifuged. Lysates are transferred to a 96-well microplate containing MagAttract magnetic particles, and genomic DNA selectively binds to the surface of the particles (see " MagAttract 96 DNA Plant Core procedure"). After washing, pure genomic DNA is eluted with low-salt buffer, and is ready for use in downstream applications. The MagAttract 96 DNA Plant Kit is designed for use with the BioRobot Plant Science system for fully automated DNA purification. QIAGEN also offers an automated protocol using the BioRobot RapidPlate workstation, and a manual protocol for evaluation purposes.
See figures

The MagAttract 96 DNA Plant Kit provides high yields of DNA from many plant species. The purified DNA is up to 40 kb in size, with fragments of 23 kb predominating, and is ready to use in downstream applications, including:

  • PCR
  • Real-time PCR
  • Microsatellite analysis
  • SNP genotyping

Are there important considerations for plasma generation and urine handling?

It is strongly advised to follow the recommendations for preparing sample material provided in the corresponding Protocol Sheet to ensure reliable results.

Plasma: It is recommended to perform plasma separation immediately after blood collection when using EDTA or citrate as anticoagulant to prevent the release of genomic DNA into the plasma fraction.

Urine: Because circulating cell-free DNA in non-stabilized urine samples is rapidly degraded after sample collection due to high nuclease activity, eluates may contain no DNA or exhibit low DNA concentration. Therefore, it is recommended to stabilize urine samples. Even when using stabilized urine, it is recommended to perform a centrifugation step immediately after stabilization to prevent the release of genomic DNA from cells. Alternatively, non-stabilized urine samples can be processed immediately after collection and centrifugation using ATL-pretreatment and automated DNA extraction as described in the corresponding Protocol Sheet.
