Biodensity of a soil or other environmental sample is not a problem when using the DNeasy PowerMax Soil Kit. This kit, designed for processing large sample quantities, will give researchers DNA samples free from PCR inhibiting compounds generally associated with soil DNA. Humic substances and the color from compost, manure and sediment are eliminated through Inhibitor Removal Technology. Resulting DNA is ready to use in a variety of downstream applications such as qPCR, Sanger sequencing and next-generation sequencing.

The DNeasy PowerMax Soil Kit was used to isolate total genomic DNA from 8 different soil samples. Isolated DNA (0.75 µg of each) was ethidium stained and analyzed on a 1% TAE agarose gel. M = Marker DNA.

PCR was used to amplify 1 µl of total genomic DNA isolated using primers to the Bacillus spp. Analysis on the amplified DNA used 0.8% TAE agarose and ethidium staining. Quantity of PCR fragmentation in each sample is relative to that in the template sequence in the purified DNA. Genomic DNA from different sample sources contains variability due to types and quantity of organisms in the respective soils. N = Negative control lacking template. P = Positive control template.

One µl was amplified using primers to the Streptococcus spp. 1% TAE agarose and ethidium staining was used to analyze the DNA. Quantity of PCR fragmentation in each sample is relative to that in the template sequence in the purified DNA.
