Optimized ELISA conditions enable the same development or incubation time.

Similar standard curves are observed for all optimized cytokine and chemokine assays in the Multi-Analyte ELISArray Kits. All standard curves are virtually superimposable indicating that all 12 assays provide similar linearity and sensitivity under the same standardized conditions and development or incubation times.

Patterns of Th1/Th2 cytokine induction.

Th1/Th2 cytokines were induced by human peripheral blood mononuclear cells in response to PMA (50 µg/ml) and ionomycin (1 µg/ml). Time-dependent patterns of induced cytokines and relative cytokine amounts were profiled and plotted.

A Multi-Analyte ELISArray Kit was used to profile multiple cytokines secreted by stimulated human peripheral blood mononuclear cells (PBMCs). The time-dependent (0, 6, 18, 24, and 48 hours) patterns of 8 Th1/Th2 cytokine inductions by human PBMCs were montored in response to PMA (50 µg/ml) and ionomycin (1 µg/ml) (see figure "Patterns of Th1/Th2 cytokine induction"). The relative amount of each of the 8 cytokines was also profiled using the Multi-Analyte ELISArray Kit.

Large amounts of IL-2 and IFNγ, and to a lesser extent TNFα, are induced throughout, and apparently beyond, the time course tested. Only more moderate amounts of IL-4 and IL-5 are induced, reaching a maximum at approximately 24 hours of stimulation. The even lower levels of IL-10 and IL-13 that are secreted also reach a maximum at 24 hours and appear to decrease again at later times. IL-12 is not detectable after 48 hours of constant stimulation.


The Custom Mix-n-Match Multi-Analyte ELISArray Kit profiles a customized panel of up to 12 cytokines or chemokines in 6 cell supernatant (media), serum, or plasma samples using one simple protocol and one standardized development time (see figure "Multi-Analyte ELISArray plate layout"). Custom Mix-n-Match Multi-Analyte ELISArray Kits provide researchers with a rapid assessment of the level of multiple proteins of their choice (cytokines and chemokines) in multiple samples.

Commercially available capture and detection antibodies are screened to achieve the highest possible sensitivity and linearity for each cytokine or chemokine assay. All assays are optimized under uniform conditions to allow simultaneous detection without compromising performance (see figure "Optimized ELISA conditions enable the same development or incubation time").


Custom Mix-n-Match Multi-Analyte ELISArray Kits use a standard sandwich ELISA protocol (see flowchart "ELISArray Protocol"). There is no requirement for special equipment; kits are compatible with a standard ELISA plate reader.
