Increased DNA yield with ClearMag technology that removes organic inhibitors
Isolate microbial DNA from soil and environmental samples with the MagAttract PowerSoil DNA Kit. Optimized for automated systems, the MagAttract PowerSoil DNA Kit processes up to 0.25 g of soil, sediment, compost or stool. The combination of Inhibitor Removal Technology and ClearMag magnetic particle technology enables the purification of DNA without organic inhibitors, providing high-quality, pure DNA for downstream applications.
Protocols are available for the Eppendorf epMotion and Thermo Scientific KingFisher, but this kit can also be used with a host of automated processing and liquid handling systems.
The MagAttract PowerSoil DNA Kit was formerly sold by MO BIO as PowerMag Soil DNA Isolation Kit.
The MagAttract PowerSoil DNA Kit is intended for molecular biology applications. This product is not intended for the diagnosis, prevention, or treatment of a disease.
Figure 1. Clean soil DNA means successful amplification.
Organic garden soil samples were processed using MagAttract PowerSoil DNA Kit with and without the Inhibitor Removal Technology (IRT) step. Both were purified using the KingFisherTM Duo. Without the IRT step, DNA failed to amplify, while those that had been processed to remove inhibitors successfully amplified and were within the standard curve (blue lines).
Figure 2. More high molecular weight DNA.
Using the MagAttract PowerSoil DNA Kit on a KingFisherTM Duo, DNA was isolated from a panel of six soil samples (0.25 g). Yields varied based on the microbial load of each sample, however high-quality, high-molecular weight DNA was isolated from each sample. Replicate samples were equal in yield and quality.
Starting samples of (a) organic garden soil, (b) low biomass soil and (c) landscaping soil are known to be high in PCR inhibitors. qPCR run on 1 µl of DNA from each of these samples shows inhibitors have been removed. Undiluted and 1:10 diluted DNA samples fell within the standard curve (grey lines) with a 3-cycle difference between diluted and undiluted samples.
