Reproducible purification of DNA from sheep ear tissue.
Sheep ear tissue samples were treated with 180 µl Buffer ATL and 20 µl QIAGEN Proteinase K at 56°C overnight. Genomic DNA was purified from the lysed tissue samples using the BioSprint 15 DNA Blood Kit with the BioSprint 15 DNA Tissue protocol. DNA was eluted in 200 µl elution buffer. Eluates (5 µl) were run on a 0.8% agarose gel in 1x TBE. M: markers (Lambda HindIII).
High PCR performance of DNA purified using the BioSprint 15 DNA Blood Kit.
Human blood was collected and treated with one of 3 anticoagulants: citrate (C), EDTA (E), or heparin (H). DNA was purified from 200 µl blood immediately after blood collection (Fresh) and after one cycle of freezing and thawing (Frozen). DNA was eluted in 200 µl elution buffer. Eluates (1 µl and 10 µl) were subjected to PCR (50 µl reaction volume) using primers to amplify a 1 kb fragment of the Hugl gene. PCR products were run on a 1.5% agarose gel in 1x TBE. M: markers.

The BioSprint 15 DNA Blood Kit is used with the BioSprint 15. The kit uses magnetic-particle technology for rapid DNA purification from cells, tissue, and blood or blood-related products. The BioSprint 15 controls an array of magnetic rods that can attract or release magnetic particles and transfer them from tube to tube. The sequential transfer of magnetic particles allows a rapid purification procedure to be performed, from the initial binding of DNA to the particles, through to washing of the particles and elution of pure DNA (see figure "Efficient processing of magnetic particles"). Since the instrument transfers magnetic particles instead of liquids, it uses minimal amounts of reagents, enabling cost-efficient sample preparation.
Procedure
Genomic DNA can be purified from cells, tissue, and blood. Lysed samples are loaded into the instrument, which then performs all steps until pure DNA is obtained (see figure "BioSprint DNA procedure"). The kit provides all the necessary reagents and plasticware for sample preparation, and is available in different sizes, allowing cost-efficient use of consumables.
For purification of genomic DNA from tissue, dried blood spots, or buccal swabs, Buffer ATL and QIAGEN Proteinase K are required (not supplied with the BioSprint 15 DNA Blood Kit).
