Product Details

The MagAttract Viral RNA Kit combines the speed and efficiency of silica-based nucleic acid purification with the convenient handling of magnetic particles. A well-established technology is used for viral RNA preparation in a 96-well format. The purification procedure is designed to ensure reproducible handling and comprises four steps: lyse, bind, wash and elute.

Principle

The MagAttract Viral RNA Kit provides fully automated purification of viral RNA from respiratory swab samples. MagAttract technology combines the speed and efficiency of silica-based RNA purification with the convenient handling of magnetic particles. Samples are processed in the KingFisher Flex Workdeck. No phenol–chloroform or manual column extraction is required.

Procedure

In the automated workflow, viral RNA binds specifically to the magnetic beads while contaminants such as divalent cations and proteins, are completely removed in three efficient wash steps. High-quality RNA is eluted in a special RNase-free buffer, ready for direct use or safe storage. The purified RNA is free of proteins, nucleases, and other contaminants or inhibitors and can be used immediately in demanding downstream applications, including PCR, qPCR, dPCR and next-generation sequencing. Protocols are available for use on the Thermo Scientific KingFisher Flex. This kit can also be used with many other automated handling systems.

Carrier RNA

Carrier RNA serves two purposes: First, it enhances binding of viral nucleic acids to the magnetic beads, especially if there are very few target molecules in the sample. Secondly, the addition of large amounts of carrier RNA reduces the chance of viral RNA degradation in the rare event that RNase molecules escape denaturation. Not adding carrier RNA to Buffer ACL may lead to reduced viral RNA recovery.

Applications

The high-quality RNA obtained using the MagAttract Viral RNA Kit and the Thermo Scientific KingFisher Flex platform is suited for use in many downstream applications, such as:

  • Viral epidemiology and genotyping
  • Infectious disease research

Who can help if it is not clear if issues are caused by the third party instrument or by the chemistry?

Please contact QIAGEN Technical Services.

3802
Who can help in case of issues with the third party instrument?

Please contact the respective QIAGEN Technical Services.

3799
Is the virus inactivated during the procedure?

This cannot be proven with the methods we have at our disposal.

3800
I would like to use the kit on another third party instrument. Who should I contact?

Please contact QIAGEN Technical Services.

3803
Can I use sample volumes other than 300 µl?

Using the same protocol, 200, 300, and 400 µl can be prepared, with the expected loss or gain in sensitivity.

3801
Are protocols also available for other instruments (e.g., Hamilton Star and Tecan Fluent)?

Not yet, but these might be added here in the future. Please contact QIAGEN Technical Services.
