Performance
pigtype Salmonella Ab is specific for the detection of antibodies to Salmonella spp. Tests were run using the pigtype Salmonella Ab Kit in the 2011 QS* ring trial. Ten serum and 10 meat juice samples were tested. All positive samples were correctly scored as being positive for salmonella antibodies. Principle
pigtype Salmonella Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with inactivated salmonella antigen. During the sample incubation, salmonella-specific antibodies bind to the immobilized antigen. The anti-IgG-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of anti-Salmonella antibodies in the sample.
Procedure
Serum, plasma, and meat juice samples can be tested after dilution. Samples are incubated for one hour with the test plate and if antibodies to Salmonella spp. are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing. The anti-IgG-HRP conjugate is added. It contains an HRP-labeled, anti-IgG-specific monoclonal antibody. Unbound conjugate is rinsed out. The color reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The results are easy to interpret.
