Principle
PyroMark Q96 Tests はPyroMark Q96 ID Systems と組み合わせて使用することで、パイロシークエンス法による正確で定量的な変異（あるいはSNP）およびメチル化解析を実現します。 これらのアッセイはPCR用プライマーとシークエンシング用プライマーで構成されています。
Procedure
パイロシークエンス法を実施する前に、ビオチン標識されたPCR産物が産生されます。ビオチン標識されたPCR 産物はストレプトアビジンでコーティングしたセファロースビーズに結合します。これらのビーズはその後PyroMark Vacuum Workstation 上のVacuum Prep Tool により捕集され、十分に洗浄された後、シークエンシング用に最適な一本鎖DNA が調製されます。PyroMark Q96 Tests はパイロシークエンス法によるメチル化解析用にデザイン済みのアッセイです。各アッセイにはビオチン標識PCR産物の作製に必要なプライマーおよびパイロシークエンス解析用のプライマーが含まれています。
