pigtype Trichinella Ab is a sensitive and specific ELISA for detecting antibodies to Trichinella spp. in serum, plasma, and meat juice samples from pigs and wild boar. Individual samples and pooled (up to 10 samples) serum and plasma samples are suitable for testing.

Performance
pigtype Trichinella Ab is specific for the detection of antibodies to Trichinella spp. A negative population of 2044 serum samples was tested. All of the samples tested negative for the Trichinella-specific antibodies, giving a diagnostic specificity 100% (see the figure Specificity of pigtype Trichinella Ab). Ten Trichinella-positive reference serum samples of the German NRL (National Reference Laboratory for Trichinella at Federal Institute for Risk Assessment, Berlin) were analyzed with the pigtype Trichinella Ab (see the table Sensitivity of the pigtype Trichinella Ab with NRL samples). In addition, serum samples of a temporal study with 2 pigs experimentally infected with T. spiralis were simultaneously analyzed. All of the serum samples tested positive 4 weeks-post-infection, giving a high diagnostic sensitivity (see the figure Sensitivity of pigtype Trichinella Ab).
Sensitivity of the pigtype Trichinella Ab with NRL samples
Ten Trichinella-positive samples of the German NRL were tested with pigtype Trichinella Ab. Samples were processed using both the short and the overnight sample incubation period. An S/P (ratio sample/positive control) ≥ 0.3 (short protocol) or ≥ 0.45 (overnight protocol) indicates the sample is positive for antibodies. All of the samples were detected positive using both incubation periods.
Principle
Nematodes of the genus Trichinella can cause Trichinellosis, a zoonosis which can cause mild to lethal illness. Humans can be infected by eating meat containing Trichinella larvae. Most pigs infected with the nematode do not show clinical signs. To prevent human infections, all pigs slaughtered for human consumption are tested by artificial digestion.
The European Commission Regulation (EC) No. 2075/2005 allows holdings to be certified as Trichinella-free under certain conditions. One such condition is a monitoring program. Serological monitoring for Trichinella-antibodies is possible and is an alternative to the cost-intensive digestion method. pigtype Trichinella Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with inactivated Trichinella antigen (E/S antigen). During the sample incubation, Trichinella-specific antibodies bind to the immobilized antigen. The anti-IgG-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of anti-Trichinella antibodies in the sample.
Procedure
Serum, plasma, and meat juice samples can be tested after dilution. There are two protocols for this product. They differ in the length of the incubation period. Samples are incubated for one hour or overnight with the test plate and if antibodies to Trichinella spp. are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing. The anti-IgG-HRP conjugate is added. It contains an HRP-labeled, anti-IgG-specific polyclonal antibody. Unbound conjugate is rinsed out. The color reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The ratio sample OD to Positive Control OD is calculated from the OD values obtained with the test sample and the Negative Control, which contains no Trichinella-specific antibodies. The values are easy to interpret.
