The DNeasy PowerPlant Pro Kit uses mechanical bead beating technology to recover high-quality DNA from tough sample times including cotton leaf and seeds, strawberry leaf, and pine needles without time-consuming or chemical laden methods such as CTAB, chloroform or phenol extraction. Pure isolated DNA is ready for downstream applications, including qPCR, Sanger sequencing and next-generation sequencing through Inhibitor Removal Technology.An optional Phenolic Separation Solution step helps provide pure DNA even from samples with high polyphenolic compounds, such as pine or grape leaves by breaking the bonds between DNA and phenolics. This step prevents DNA loss that would otherwise occur in these samples.

Use the DNeasy PowerPlant Pro Kit in concert with vortex or high velocity bead beating, based on sample needs. Vortex methods work with soft leaf tissue, while high velocity bead beaters, like the PowerLyzer 24 Homogenizer, will break down tougher plant material such as roots, seeds, stems or difficult leaf tissues.

Purification of DNA using the DNeasy PowerPlant Pro Kit can be automated on the QIAcube Connect.

The DNeasy PowerPlant Pro Kit isolates high-quality genomic DNA from plant samples. Using a 1% TAE agarose gel, DNA was isolated from grass (2 µl), strawberry leaf (15 µl), grape leaf (15 µl) and pine needles (1 µl).

Using the DNeasy PowerPlant Pro Kit and plant DNA isolation kits from three other suppliers, DNA was isolated from cotton seeds. Isolations were performed per each manufacturer’s protocols. A 1% TAE agarose gel was used to examine a 2% elution of DNA volume.

Using RuBisCo primers run through qPCR on 1 ng of DNA from pine needles (A) and strawberry leaf (B), both diluted 1:10 and undiluted DNA samples fell within the standard curve (purple lines). The difference between the diluted and undiluted samples was approximately 3 cycles indicating that the samples were freed of PCR inhibitors.
