Specifications

  • Key features

The QIAxpert is reduced to the max! The very compact system offers maximum convenience through simple sample handling and innovative software features.

  • Microfluidic features of the sample carrier

Expert design of the QIAxpert Slide ensures clean sample loading, flexible use of sample positions and preservation of the samples for up to 2 hours.

  • Spectral profiling analysis

Beyond UV/VIS quantification at 260 and 280 nm wavelengths, QIAxpert Software performs spectral decomposition to selectively quantify DNA, RNA or protein in a sample.
[A] A highly pure sample of RNA shows no significant contamination as evidenced by the simple spectrum. [B] A sample of RNA was spiked with gDNA. The RNA is indicated by the blue absorbance line while the spiked gDNA generates the red absorbance line.

  • Report and Export

QIAxpert automatically generates comprehensive reports that can be viewed in any browser. Graphs can be extracted from these HTML files via drag and drop. Additionally, tabular data are available in .csv and .txt formats. To facilitate data analysis and sharing, the data are exportable to a USB stick, a network drive or a smart device via a QR code.

Analysis protocols

QIAxpert performs classic nucleic acid and protein quantification based on absorbance measurements. In addition, smart analysis protocols called “apps” come pre-installed to quantify specific analytes of interest. Based on top bioinformatics expertise, these spectral profiling protocols let users focus on the molecule of interest, subtracting other absorbing impurities and background turbidity.

Sample carrier

Watch the video to learn about the benefits provided by the microfluidic QIAxpert slides. Along with minimal sample volume requirements, no cleaning is needed between runs, and there is no risk of cross-contamination.

Operation

Watch how simple it is to operate the QIAxpert. Samples are easily loaded, and results are displayed in less than 2 minutes.

Reproducibility

The QIAxpert provides highly reproducible results over a wide dynamic range. In this experiment, 5 replicates of each sample from a dilution series of salmon sperm DNA were measured on the QIAxpert. Reproducibility was assessed by calculating the coefficient of variation.

Linearity

To assess the linearity of the system, pure gDNA was serially diluted to generate samples from 2000 to 2 ng. The resulting dilutions were then measured on the QIAxpert using 2 µl for each of 5 replicates. The quantifications from the QIAxpert were plotted against the known concentration values. The resulting plot demonstrates the exceptional linearity (with a regression coefficient of >0.999) of QIAxpert up to 2000 ng/ml or 40 OD (A260, 10 mm).

Spectral content profiling

Quantification of dsDNA purified with QIAamp silica membrane technology is shown. The concentration was determined based on the absorbance at 260 nm and using a factor of 50.
The blue curve shows the DNA portion measured. Other absorbing impurities and nonDNA nucleic acids are subtracted from the result (orange curve) and the background is checked for turbidities (none in this example), which would be represented as a gray curve. The yellow line is the residual spectrum, which is the part of the measured spectrum that cannot be attributed to the reference profiles used in the algorithm.

UV/VIS scan

Results for dsDNA measured with the A260 dsDNA app are shown. This app quantifies dsDNA based on the total absorbance at 260 nm using the extinction coefficient for dsDNA of 50 µl/ng*cm.

UV/VIS scan

Results for total RNA measured with the A260 RNA app are shown. This app quantifies total RNA based on the total absorbance at 260 nm using the molar extinction coefficient for RNA of 40 µl/ng*cm.

Spectral content profiling

Results of quantification of total RNA purified with RNeasy technology are shown. The concentration was determined based on the absorbance at 260 nm using the molar extinction coefficient for RNA of 40 µl/ng*cm.

The blue curve shows the RNA portion measured. Other absorbing impurities and nonRNA nucleic acids are subtracted from the result (orange curve) and the background is checked for turbidities (gray curve). The yellow line is the residual spectrum, which is the part of the measured spectrum that cannot be attributed to the reference profiles used in the algorithm.

Protein

Results for purified protein measured with the A280 Protein app are shown. This app quantifies purified protein based on the total absorbance at 280 nm and using extinction coefficients entered prior to result analysis.
