Principle
miScript Inhibitor Negative Control is a chemically synthesized, single-stranded, modified RNA designed to target the sequence of AllStars Negative Control siRNA. miScript Inhibitor Negative Control has no homology to any known mammalian gene. It is recommended for use as a negative control in every miRNA inhibitor experiment.

A negative control should be transfected in every miRNA inhibitor experiment and will indicate if results are nonspecific. Comparison of results from the negative control with results from the inhibitor under study can be used to confirm that the observed results are specific to the inhibitor under study. miScript Inhibitor Negative Control targets the sequence of AllStars Negative Control siRNA, the most thoroughly tested small RNA control currently available.

Procedure
In experiments in which an endogenous gene will be used for downstream analysis, the negative control should be a transfection of miScript Inhibitor Negative Control. In experiments in which a reporter vector will be used for downstream analysis, the negative control should be a cotransfection of miScript Inhibitor Negative Control and the reporter vector. Futher details on miRNA inhibitor experiments and controls are available in the handbook Guidelines for miRNA mimic and miRNA inhibitor experiments.
Applications

A negative control should be transfected in every miRNA inhibitor experiment to confirm that the observed results are specific to the inhibitor under study. miScript Inhibitor Negative Control can be used in experiments in:

miRNA research
Gene regulation research
