Blood was collected from 12 donors into PAXgene Blood RNA Tubes. RNA was purified using the QIAsymphony PAXgene Blood RNA Kit on the QIAsymphony SP. RNA was analyzed with an Agilent 2100 Bioanalyzer. Performance

全自動でのPAXgene Blood RNA 法はマニュアルでのPAXgene Blood RNA 法に相当するRNA収量を実現します。2.5 mlの健常なヒトの全血（4.6 x 106 ～1.1 x 107 白血球/ml）から分離されるRNAの一般的な収量は、すべてのPAXgene Blood RNA Kitsで処理した検体の95％以上で3 µg 以上です。PAXgene Blood RNAの自動調製は様々なドナー検体から豊富なsmall RNA種を含む高品質RNA精製を実現します（図“様々なドナーからの高品質RNA”）。収量はドナー依存性が高いため、個人個人での収量は変動します。また、DNase処理が含まれているので、ゲノムDNAの混入は最小限に抑えられます。精製したRNA はリアルタイム定量RT-PCRのようなダウンストリームアプリケーションに直接使用できます。

Principle

QIAsymphony PAXgene Blood RNA Kitは、QIAsymphony SP上で細胞RNA精製の完全自動化を実現します。サンプル採取と安定化用にパクスジーンRNA採血管（日本ベクトンｷディッキンソン株式会社より入手可能、cat. no. 762165）を使用すると、本システムは採血と安定化から実証済みの磁性粒子テクノロジーを用いた高品質RNA精製まで、プレアナリティカルのトータルソリューションを実現します。画期的な充填済みカートリッジはマニュアルでの作業時間の大幅な削減を実現し、さらにバーコードによる試薬の使用履歴のトラッキングもできます。

Procedure
QIAsymphony PAXgene Blood RNA調製はパクスジーンRNA採血管に血液を採取（2.5 ml）することから開始します。サンプルは室温で保存あるいは輸送できます。精製法は、まず遠心操作によりパクスジーンRNA採血管の内容物をペレット化します。ペレットを再懸濁した後、チューブをQIAsymphony SPに移します。試薬カートリッジおよびプラスチック容器の設置は専用ソフトウェアにより指示、チェックされます。その後、QIAsymphony SPによるRNAの自動精製が開始します。DNase処理が含まれているのでゲノムDNAの混入は最小限に抑えられます（図“QIAsymphony PAXgene Blood RNA操作”）。適応性が高く標準化された調製法はQIAsymphony SPのオンボード対策との組み合わせでサンプル間のクロスコンタミが最小限に抑えられるようにデザインされています（図“クロスコンタミに対するオンボード対策”）。精製後、溶出液の最終熱処理によりダウンストリームアプリケーションでのパフォーマンスが高まります。所要時間は24サンプルで2.5時間、実際のマニュアルでの作業時間は最大25～30分です。
