The QIAamp DNA Host-Free Microbiome Kit selectively isolates microbial DNA from samples rich in host DNA, such as tissues, bodily fluids and swabs, promoting accurate microbiome analysis due to efficient host nucleic acid depletion. Furthermore, this kit extracts high-yield bacterial DNA, ideal for metagenomic sequencing and suited for qPCR, dPCR and other molecular applications.
The QIAamp DNA Host-Free Microbiome Kit depletes host DNA while preserving microbial DNA integrity (Figure 1). Isolated microbial DNA is high-yield and largely free from host contamination (Figure 2). Mechanical and chemical disruption methods enhance lysis and ensure high DNA recovery (Figure 3). The resulting DNA is of high purity and can be used for various downstream applications, including next-generation sequencing and PCR-based assays.
Microbes coevolved with humans and other animals as an integral part of health. However, they also play roles in disease. Changes in the microbiome, including a loss of e.g., bacterial diversity (dysbiosis) contribute to inflammatory and tumorous diseases.
Direct 16S rRNA sequencing analyzes bacterial taxonomic profiles and the relationships between dysbiosis and various diseases. However, tissues, swabs and bodily fluids contain multitudes more host DNA than bacterial DNA. This host DNA contamination lowers the analysis efficiency and costs valuable reads in metagenomic sequencing studies. QIAamp DNA Host-Free Microbiome Kit overcomes this imbalance by removing host DNA. QIAamp DNA Host-Free Microbiome Kit is ideal for metagenomic sequencing and supports applications such as qPCR, digital PCR, and 16S rRNA gene sequencing for comprehensive microbial profiling.
Host nucleic acids are selectively degraded before microbial DNA isolation. During depletion, the bacterial cells remain intact, which results in highly specific microbial DNA isolation. DNA purification is then performed using spin columns, ensuring efficient binding and elution.
Samples are treated with Host Depletion Solution to digest host nucleic acids while keeping bacterial cells intact. Bacterial cells are then disrupted using PowerBead Pro Tubes and lysis buffer. The lysates are transferred to QIAamp ultra-clean production Mini Columns, where microbial nucleic acids are adsorbed onto silica membranes. Two wash buffers remove contaminants, ensuring high-purity DNA. Finally, the purified bacterial DNA is eluted with RNase-free water, ready for downstream applications.
The QIAamp DNA Host-Free Microbiome Kit is ideal for microbiome research, metagenomics and infectious disease studies. It supports sensitive downstream applications and is ideal for enhancing reads in metagenomics studies. Host-depleted DNA is also suitable for qPCR, digital PCR and 16S rRNA gene sequencing.
DNA was isolated from bovine omasum using the QIAamp DNA Host-Free Microbiome Kit and the predecessor QIAamp DNA Microbiome Kit. The QIAamp DNA Host-Free Microbiome Kit efficiently depletes host DNA while extracting approximately 6 times more bacterial DNA. Total DNA was measured by using fluorometric quantification (Qubit™). Bacterial DNA was quantified by 16S rRNA qPCR using QuantiNova SYBR Green PCR Kit. Error represents standard deviation of quadruplicates.
