AllPrep RNA/Protein Kitは同一の細胞培養液サンプルからトータルRNAとネイティブなタンパク質を同時に分離精製します。細胞サンプルから精製したRNAをAgilent 2100 Bioanalyzerで解析すると、RIN値は通常10となり、RNAが分解されていないことが示されます。同一の細胞サンプルから精製されたネイティブなタンパク質には、全ての細胞タンパク質が含まれています（図“ 同一の細胞サンプルから精製された全く分解されていない高品質なRNAおよびタンパク質を精製"）。AllPrep RNA/Protein Kitにより精製されたRNAはリアルタイムRT-PCRに適しており（図" リアルタイムRT-PCRにおける感度"）、精製された細胞タンパク質はダウンストリームの酵素アッセイにおいて機能性が示されています（図" 機能タンパク質の精製"）。

Principle
AllPrep RNA/Protein Kitは同一の細胞培養液サンプルからトータルRNAとネイティブなタンパク質を同時に分離精製します。mRNAとタンパク質のレベルでsiRNAによるノックダウンを検証する実験において、本キットはサンプル調製を合理化するとともに、培養細胞数を顕著に減らせます。また、RNAおよびタンパク質は同一サンプル由来であるため、ノックダウン解析の信頼性がより高くなります。AllPrep RNA/Protein Kitは、RNAiおよび遺伝子発現解析用に開発されたQIAGEN社の製品グループの一部です。これらの製品によって、再現性が極めて高い、高感度な結果が得られます。これらの製品グループには、至適化済みのPCRマスターミックス、即使用できるアッセイ、siRNA、トランスフェクション試薬が含まれています。これらの製品を用いたRNAiや遺伝子発現解析のワークフローの標準化は、バイオメディカル研究の多くの分野での成功に寄与しています。
Procedure

Highly intact RNA and high-quality protein from the same cell sample.
Total RNA was purified from HeLa cells and analyzed on the Agilent 2100 Bioanalyzer. The rRNA ratio (28S/18S) was 2.04. Total protein was purified from HeLa cells and analyzed by western blot. Analysis of duplicate protein samples with antibodies against Lamin (numclear protein) and TIM23 (mitochondrial protein) demonstrates that the total protein fraction contains proteins from subcellular compartments.
