QIAxcel instruments, when used in conjunction with QIAxcel DNA Kits, provide fully automated size separation and quantification of DNA fragments in up to 96 samples per run. QIAxcel technology, based on capillary electrophoresis using gel cartridges, provides unmatched resolution, speed, and throughput. QIAxcel gel cartridges are reusable, allowing up to 250 runs of 12 samples to be performed. Preinstalled methods suitable for most applications are provided. QIAGEN can also create customized methods for users.

​The QIAxcel DNA Kits are only used in combination with the QIAxcel Advanced System and the QIAxcel DNA Accessories.

Performance

Easy high-resolution analysis
Genotyping organisms, PCR-based cDNA library screening, and many other biomedical research and molecular biology applications require discrimination of PCR fragments that vary in size by a small number of base pairs. The QIAxcel High Resolution Kit, in combination with QIAxcel instruments, allows analysis of DNA fragments between 15 bp and 5 kb in size. Fragments smaller than 500 bp can be separated with a resolution of 3–5 bp, providing greater confidence in data interpretation than is possible with conventional high-resolution agarose gel electrophoresis (see figure  Sensitive detection of a mutated cancer-related gene). At a detection sensitivity of 0.1 ng/µl DNA in undiluted amplification reactions, QIAxcel instruments offer more robust results with less sample input material.
 
The QIAxcel DNA Screening Kit is highly suited for routine evaluation of fragments for restriction endonuclease digestion, qualitative singleplex or multiplex PCR, and other DNA applications (see figure  High-resolution analysis of multiplex PCR samples). The use of gel cartridges for DNA screening streamlines the lab workflow by minimizing analysis time for fragments of 15 bp to 5 kb in size - 96 samples are processed in approximately 1 hour. A resolution of 20–50 bp can be obtained for fragments smaller than 1 kb. Sample consumption is less than 0.1 µl, leaving virtually all of your precious sample for further downstream use.
 
The QIAxcel DNA Fast Analysis Kit is highly suited for routine evaluation of DNA fragments in qualitative single or multiplex PCR applications. Use of gel cartridges for DNA screening streamlines the lab workflow by minimizing analysis time for fragments of 15 bp to 3 kb in size — 12 samples are processed in just 3 minutes and 96 samples in approximately 25 minutes (see figure  Screenshot showing data from PCR fragment analysis). A resolution of down to 50 bp can be achieved with the QIAxcel DNA Fast Analysis gel cartridge, while sample consumption is less than 0.1 μl.

Principle

QIAxcel instruments use capillary gel electrophoresis to enable fast size-based separation of nucleic acids (see figure " Sample separation process"). Unlike traditional agarose gel electrophoresis, separation is performed in a capillary of a ready-to-use gel cartridge. The samples are automatically loaded into an individual capillary and voltage is applied. The negatively charged nucleic acid molecules migrate through the capillary to the positively charged terminus. As with agarose gel electrophoresis, low-molecular-weight molecules migrate faster than high-molecular-weight molecules. As the molecules migrate through the capillary, they pass a detector which detects and measures the fluorescent signal. A photomultiplier converts the fluorescent signal into electronic data, which are then transferred to the computer for further processing using QIAxcel ScreenGel software. After processing, the data are displayed as an electropherogram or gel image.

Procedure

QIAxcel DNA Kits simplify separation and analysis of DNA fragments, and provide increased user safety. Just a few simple steps are required: load the gel cartridge, fill and load the buffer tray, load the samples in 96-well plates or in PCR tubes or strips, select the process profile to be used - and go! Tedious gel preparation and extensive user training are eliminated, streamlining the workflow and facilitating integration of the system into the daily lab routine. Within minutes of starting a run, the first results appear in real time on the computer screen (see figure " Reduced run times").

Applications

Preprogrammed methods, in combination with the corresponding gel cartridges, allow separation and analysis of a variety of nucleic acids, including single or multiple PCR fragments, DNA digested with restriction endonucleases, and synthesized oligonucleotides.

FAQ

Why does an intensity calibration of the gel cartridge for the QIAxcel System and QIAxcel Advanced need to be performed?

The intensity calibration procedure on the QIAxcel System and QIAxcel Advanced is performed to normalize the signal intensities across all 12 channels of the gel cartridge. This corrects for natural intensity reading variations between each capillary in the cartridge. A correction factor is determined and applied for every subsequent run performed with the cartridge.

Can a QIAxcel DNA Fast Analysis Cartridge be run on a QIAxcel System?

The QIAxcel DNA Fast Analysis Cartridge can only be run on the QIAxcel Fast Analysis System with the BioCalculator Fast Analysis software and the grey software key.

All other QIAxcel Cartridges can only be run on the QIAxcel System with the BioCalculator software and the blue software key.

Can customers use their own DNA size marker with the QIAxcel System and if yes, what concentration should be used?

The concentration of the DNA marker, pUC18/HaeIII, provided in the QIAxcel DNA Kits is 5ug/ml in 1XTE buffer solution. Customers can run their own DNA size marker sample with different concentration levels using the provided Methods (i.e. M500, H500, L500 and so on).

The recommended method for all basic applications with the QIAxcel System is the M500 method. The BioCalculator software also allows customers to choose their own Sample Injection Time in the Sequence table. The M500 method provides 5KV for 20 seconds for the electro-kinetic injection of lower concentrated DNA samples. If the Injection Time is doubled or tripled (40 or 60 sec), you can expect double or triple intensities (also peak heights).

If a different DNA size and/or alignment marker is to be used with the QIAxcel System or QIAxcel Advanced, does the Gel Cartridge need to be recalibrated?

The Gel Cartridge does not need to be recalibrated when using a new alignment marker and/or DNA size marker with the QIAxcel System or QIAxcel Advanced. Calibration is required only once, i.e. when the first cartridge is run or when the cartridge is used on a different QIAxcel system than the one it has been calibrated on, or when using a different PC than the one used to calibrate initially.

How can drying out of the gel cartridge tips used on the QIAxcel System and QIAxcel Advanced be prevented?

If the gel cartridge tips for the QIAxcel System and QIAxcel Advanced are in contact with air, the tips will dry out. There are 3 ways of preventing the tips from drying.

  1. Store the cartridge in the instrument with the instrument in the “Park” position. The washing solution (8 ml) covered with mineral oil must be in the “Park” position of the Solution Tray. 
  2. Store the cartridge in the Cartridge Stand with enough mineral oil in the well of the Cartridge Stand to cover the capillary tips. The Cartridge Stand should only be used for short term storage, i.e. when changing cartridges during one working day. 
  3. Place the cartridge back into its packaging with the capillary tips gently inserted into the soft gel in the box.
