This PCR assay has been validated as 100% specific for Brachyspira hyodysenteriae DNA. The validation was carried out at the Veterinary Laboratory Agency (VLA) Weybridge, UK.
Pathogen identification by PCR is based on the amplification of unique specific regions of the pathogen genome. In real-time PCR, the amplified product is detected via fluorescent dyes. These are usually linked to oligonucleotide probes that bind specifically to the amplified product. Monitoring the fluorescence intensities during the PCR run (i.e., in real time) allows the detection of the accumulating product without having to re-open the reaction tubes after the PCR run.
The cador B. hyodysenteriae PCR Reagent is a ready-to-use system for the identification of DNA from Brachyspira hyodysenteriae using PCR. The kit contains primer-probe sets for specific amplification of a highly conserved region of the Brachyspira hyodysenteriae genome. The kit also contains an additional heterologous amplification system to identify possible PCR inhibition, and an external positive control. Real-time PCR detection can be carried out on the Rotor-Gene Q thermal cycler.
Bacterial DNA can be manually isolated using the QIAamp cador Pathogen Mini Kit or the QIAamp DNA Mini Kit. Both isolation protocols are fully automatable on the QIAcube. The isolated DNA is ready for use with the cador B. hyodysenteriae PCR Reagent on the Rotor-Gene Q real-time PCR cycler.
