Performance
The DNeasy 96 Plant Kit provides high-throughput isolation of high-quality DNA from 96 or 192 samples of plant leaf tissue in less than 2 hours. The simple 96-well procedure provides highly reproducible yields of total cellular DNA (see figures "Uniform DNA yields" and "Reproducible DNA yield and quality"). The typical yield is 1–15 µg per 50 mg plant leaf tissue, with an elution volume of 100–200 µl. DNA yields vary between species depending on genome size, ploidy, cell number, and age of tissue sample. DNeasy purified DNA is sized up to 40 kb, with fragments of 20–25 kb predominating. Polysaccharides, proteins, and enzyme inhibitors are efficiently removed. Absorbance scans of DNeasy purified DNA show a symmetrical peak at 260 nm, confirming that the DNA is free of impurities, including enzyme inhibitors. Principle
The DNeasy 96 Plant Kit uses advanced silica-membrane technology and simple spin procedures to isolate highly pure total cellular DNA from plant tissues and cells or fungi. The streamlined DNeasy 96 procedure and convenient 96-well format eliminate tedious labeling, one-at-a-time pipetting, and DNA resuspension steps, and help minimize cross-contamination. DNeasy technology replaces cumbersome DNA isolation procedures such as cetyltrimethylammonium bromide (CTAB), phenol, or chloroform extraction. Using the DNeasy procedure, alcohol precipitation is not necessary — purified DNA is ready for immediate use. DNeasy sample preparation technology is fully licensed.
Procedure
Samples are first mechanically disrupted and then chemically lysed (see flowchart "DNeasy Plant procedure"). RNA is removed by RNase digestion during lysis. Cell debries, precipitated proteins and polysaccharides are removed by centrifugation. Buffering conditions are adjusted, and the lysates are loaded onto the DNAeasy Plant 96-well plate (see figure "DNeasy 96 Plant plate"). During a brief spin, DNA selectively binds to the silica membrane while contaminants pass through. Remaining contaminants and enzyme inhibitors are removed in one efficient wash step. Pure DNA is then eluted in water or low-salt buffer, ready for use. Processing of DNeasy 96-well plates requires use of the QIAGEN 96-Well-Plate Centrifugation System.
