3331 - Should the REPLI-g amplified mtDNA be cleaned up before sequencing?

NGS sequencing starts with a fragmentation step. It is not necessary to purify REPLI-g amplified mtDNA prior to DNA fragmentation because after fragmentation, a cleanup is necessary anyhow for size selection.

Sanger sequencing starts with a PCR to select a short region suitable for Sanger sequencing. For PCR, no purification of REPLI-g amplified mtDNA is necessary.
