The EZ2 AdnaTest CTC Select Kit provides immunomagnetic enrichment of circulating tumor cells from human whole blood on the EZ2 Connect instrument. Integrated mRNA isolation allows molecular characterization of CTCs using RT-PCR, real-time RT-PCR, or cDNA generation, including the AdnaTest procedure for molecular characterization of CTCs.

Performance

EZ2 AdnaTest CTC Select Kit allows the automated enrichment of CTCs from human whole blood based on the AdnaTest principle using the EZ2 Connect instrument. The EZ2 AdnaTest CTC Select Kit is a highly specific immunomagnetic cell-selection system for enriching circulating tumor cells (see Table "Performance comparable to manual protocol – EZ2 AdnaTest CTC Select specificity and recovery") from peripheral blood including integrated mRNA isolation. The EZ2 Connect automates the CTC enrichment and subsequent mRNA isolation using prefilled cartridges. The benchtop instrument automatically pierces the sealed cartridges, and automates pipetting of beads and buffers. Its intuitive touchscreen interface makes it easy to use, even for new lab workers. And with its remote connectivity feature, users can monitor runs and the instrument can be managed even from outside the lab (see figure " Easy to use – enrich circulating tumor cells for molecular analyses”).

The high-quality mRNA purified has proven identical specificity and sensitivity in downstream applications to mRNA isolated by the manual protocol, and can therefore be used in any application, such as RT-PCR, real-time RT-PCR, or cDNA generation, including the AdnaTest procedure for molecular characterization of CTCs. In combination with EMT-2 Add-on CancerDetect products, the resulting mRNA/cDNA can therefore be used to generate results for the following cancer entities:

Principle

EZ2 AdnaTest CTC Select provides prefilled cartridges containing all buffers for the enrichment and lysis of CTCs. A fresh blood sample is incubated with CTC Select beads before loading the sample into the EZ2 Connect instrument. The instrument automatically pierces the prefilled cartridges and enriches the tumor cells using in-tip magnetic bead separation. AdnaTest CTC Select enables the immunomagnetic enrichment of tumor cells from whole blood via epithelial- and tumor-associated antigens. Antibodies against epithelial- and tumor-associated antigens are conjugated to magnetic beads for labeling of tumor cells. Labeled cells are extracted by the EZ2 Connect and lysed. The cartridges also contain oligo (dT) 25-coated beads for isolation of mRNA from the lysate of pre-enriched tumor cells (see figure " During the run"). Reverse transcription results in cDNA that can be used as template for tumor cell detection and characterization by PCR.

Importantly, the AdnaTest is an open platform that allows the profiling of additional custom-selected mRNA targets for comprehensive biomarker discovery.

Procedure

Fresh blood samples are incubated with CTC Select beads before being loaded into the EZ2 Connect instrument. Oligo dT beads and an elution tube are added to the prefilled EZ2 AdnaTest CTC Select cartridges before loading together with the preincubated samples into the EZ2 Connect instrument. Instrument setup is finalized by adding the large volume rack (cat. no. 9027011) and adding tips and tip holders (provided separately, cat. no. 994900). The EZ2 AdnaTest CTC Select protocol is selected and the instrument started (see figure:  Getting started).

FAQ

Can samples other than whole blood be used with the EZ1 DSP DNA Blood Kit?
No. The EZ1 DSP DNA Blood system is a generic system that uses magnetic-particle technology for automated isolation and purification of genomic DNA from human whole blood samples, for in vitro diagnostic purposes. The system is not for use with tissue or stool samples. It is also not for isolation or purification of bacterial, fungal, viral, or parasite nucleic acids, nor for isolation or purification of RNA. The system performance has not been evaluated for isolation or purification of genomic DNA from buffy coat, cultured or isolated cells, swabs, dried blood spots, or cell-free body fluids, such as liquor, plasma, serum, or urine. Contact your local QIAGEN representative for information about other EZ1 Kits.

How long can blood samples be stored before sample preparation using the EZ1 DSP DNA Blood system?
For short-term storage (up to 7 days), collect blood in tubes containing EDTA as an anticoagulant and store the tubes at 2–8°C. However, for applications requiring maximum fragment size, such as southern blotting, we recommend storage at 2–8°C for up to 3 days only, as low levels of DNA degradation will occur after this time. 

For long-term storage, collect blood in tubes containing a standard anticoagulant (preferably EDTA, if high-molecular-weight DNA is required) and store the tubes at −20°C for a maximum of 4 weeks. Longer storage may be possible depending on the downstream application but needs to be validated by the user.

Yield and quality of the purified DNA may depend on storage conditions of the blood. Fresh blood samples may yield better results. Do not re-freeze blood samples more than 2 times as this may result in decreasing DNA yield.

Can I use sample preparation cartridges from other suppliers together with the EZ1 instruments or EZ2 Connect MDx?
No. The EZ1 instruments or the EZ2 Connect MDx are intended to be used only with QIAGEN sample preparation cartridges.

What do I have to do if precipitates are visible in the lysis buffer (well 1 of the reagent cartridge [RCB])?
Place the reagent cartridges (RCB) into a shaker–incubator, and incubate at 30–40°C with mild agitation for up to 2 hours. Do not use the reagent cartridges (RCB) if the precipitates do not redissolve.

How long can eluates be stored after sample preparation using the EZ1 instruments or EZ2 Connect MDx and the EZ1 DSP DNA Blood Kit?
We recommend storing the purified DNA at 2–8°C or at –20°C for up to 24 months. For extended storage time, we recommend storage at –20°C or –80°C for up to 36 months. The impact of DNA stability might be different for the specific downstream application being used and needs to be self-validated by the user.

How can I track samples during collection, sample preparation, and downstream assay?
A sample sheet template is included in the handbook. This sample sheet may be useful for recordkeeping when using the EZ1 DSP DNA Blood or the EZ1 DSP Virus procedure. This sheet can be photocopied and labeled with descriptions of the samples and details of the run.

How long can eluates be stored after sample preparation using the EZ1 instruments or EZ2 Connect MDx and the EZ1 DSP Virus Kit?
For short-term storage of up to 24 hours, it is recommended to store the purified viral nucleic acids or bacterial DNA at 2–8°C. For long-term storage of more than 24 hours, it is recommended to store at –80°C for up to 12 months or –20°C for up to 12 weeks. Stability of nucleic acids might be different for the specific downstream application being used and needs to be self-validated by the user.
