One example of a Multi-Analyte ELISArray Kit application is as follows. A Multi-Analyte ELISArray Kit was used to profile multiple cytokines secreted by stimulated human peripheral blood mononuclear cells (PBMCs). The time-dependent (0, 6, 18, 24, and 48 hours) patterns of 8 Th1/Th2 cytokine inductions by human PBMCs were montored in response to PMA (50 µg/ml) and ionomycin (1 µg/ml) (see figure " Patterns of Th1/Th2 cytokine induction". The relative amount of each of the 8 cytokines was also profiled using the Multi-Analyte ELISArray Kit.
Large amounts of IL-2 and IFNγ, and to a lesser extent TNFα, are induced throughout, and apparently beyond, the time course tested. Only more moderate amounts of IL4 and IL5 are induced, reaching a maximum at approximately 24 hours of stimulation. The even lower levels of IL10 and IL13 that are secreted also reach a maximum at 24 hours and appear to decrease again at later times. The amount of IL12 is not detectable after 48 hours of constant stimulation.
