The Investigator Triplex DSF Kit is for prescreening of human DNA samples and can be used as a starting point for genotyping strategies in human identity and forensic testing. It enables simultaneous amplification of STR loci D3S1358, SE33 (ACTBP2), and FGA (FIBRA) from low-copy and degraded DNA. The genetic analysis is calibrated with the DS-30 matrix.

Performance

The Investigator Triplex DSF Kit was validated and evaluated using the GeneAmp 9700 thermal cycler (in standard mode), and the ABI PRISM 310, ABI PRISM 3100, and Applied Biosystems 3130 Genetic Analyzers.

The recommended amount of DNA under standard conditions is 0.35 ng. Internal validations demonstrated robust and balanced results with 0.1–1.0 ng DNA, and reliable results with <0.1 ng DNA.

Principle

The Investigator Triplex DSF Kit enables the simultaneous amplification of the 3 polymorphic STR loci D3S1358, SE33 (ACTBP2), and FGA (FIBRA) using two flourescence label dyes. The kit includes optimized primers and reagents.

Procedure

The 3 polymorphic STR loci are amplified simultaneously and labeled with the flourescent dyes as listed in the table "Investigator Triplex DSF dyes and corresponding markers".

The amplified loci undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as QIAGEN's Investigator IDproof or IDproof Mixture Software. See the table "Technical specifications" for more information.
