The Investigator ESSplex Plus Kit amplifies the full suite of 15 STR markers in the new European Standard Set (ESS) of loci, plus the gender-specific Amelogenin, as recommended by the European Network of Forensic Science Institutes (ENFSI) and approved by the European Union Council. The kit utilizes QIAGEN’s fast cycling technology, enabling amplification in just 90 minutes. The genetic analysis is calibrated with BT5 Matrix, an optimized dye technology.

The effective run time is shown for the Investigator ESSplex Plus Kit versus products from other suppliers. The PCR was run on an Applied Biosystems GeneAmp PCR System 9700 Thermal Cycler with Gold-plated Silver block.|The electropherogram displays a profile of 500 pg Control DNA XY5 generated in the presence of 500 μM Hematin. The analysis was performed on an Applied Biosystems 3500 Genetic Analyzer using the Investigator IDproof Software.
|The assay performance was tested in the presence of increasing concentrations of 6 types of inhibitor. 500 pg of Control DNA 9948 was added to the master mix and PCR was performed under standard conditions. The analysis was performed on an Applied Biosystems 3500 Genetic Analyzer using the Investigator IDproof Software. 100 rfu was used as the threshold for allele calling.|
Performance

The Investigator ESSplex Plus Kit utilizes fast-cycling technology to provide reliable results in approximately 90 minutes, which is significantly faster than kits from other suppliers (see the figure "PCR run time comparison").

The novel Fast Reaction Mix is based on proven QIAGEN multiplex PCR technology that has been further optimized to increase inhibitor tolerance. Assay performance tested in the presence of increasing concentrations of 6 types of inhibitor (humic acid, hematin, tannic acid, indigo carmine, calcium, and collagen) shows high sensitivity and superior inhibitor resistance (see the figures "Profile generation in the presence of inhibitors" and "Assay inhibitor resistance"). This enables the generation of reliable, high-quality results.

The Investigator ESSplex Plus Kit was validated and evaluated using the GeneAmp 9700 thermal cycler (in standard mode) and the Applied Biosystems 3500 Genetic Analyzer.

The recommended amount of DNA under standard conditions is 0.5 ng. Internal validations demonstrated robust and balanced results with 0.2-2 ng DNA, and reliable results with <0.1 ng DNA.

Principle

The Investigator ESSplex Plus Kit uses QIAGEN's fast-cycling PCR technology for the rapid, simultaneous amplification of the 15 polymorphic STR markers that comprise the new European Standard Set (ESS) and Amelogenin. Carefully designed primers prevent overlap of loci alleles, thereby minimizing the likelihood of misinterpretation.

Procedure

The 15 polymorphic STR markers and the gender-specific Amelogenin are amplified simultaneously in a single PCR and labeled with one of the dyes listed in the table "Investigator ESSplex Plus Kit dyes and corresponding markers".

The amplified loci then undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as QIAGEN's Investigator IDproof or IDproof Mixture Software. See the table "Technical specifications" for more information.
