This protocol is for purification of up to 20 µg plasmid DNA (>10 kb) from 1–5 ml overnight cultures of E. coli in LB (Luria-Bertani) medium using the QIAprep Miniprep Spin Kit. The protocol includes a wash step with Buffer PB and an elution step with Buffer EB preheated to 70°C. |
