The miRCURY LNA miRNA QC PCR Panel enables quick and easy quality control of RNA samples, so you can be sure to use only optimal samples for your qPCR profiling studies. Use the 96- or 384-well format PCR panel to check the quality of your isolated RNA, look for outlier samples, identify the presence of nucleases and PCR inhibitors and look for signs of hemolysis. Avoid spending time and resources on suboptimal samples.

The former Exiqon "miRCURY LNA Universal RT microRNA PCR System" has been updated to the "miRCURY LNA miRNA PCR System" with new reagents that ensure the same superior performance:

  • The Universal cDNA Synthesis Kit II has been replaced by the miRCURY LNA RT Kit
  • The ExiLENT SYBR Green Master Mix Kit has been replaced by the miRCURY LNA SYBR Green PCR Kit
  • Most miRCURY LNA miRNA PCR Assays remain unchanged, but 15 validated assays have been redesigned; refer to the Product Resources tab for this assay list

miRCURY LNA miRNA QC PCR Panels can be used with a wide range of sample types.
The miRNAs on the panel are expressed in many sample types. In this example, human serum, plasma, urine, colorectal cancer (FFPE) and mouse brain (fresh frozen) samples were analyzed.

Determine RNA yields using the RNA Spike-in Kit.
miRCURY LNA miRNA QC PCR Panels offer a convenient way of determining RNA yields. In this example, a plasma sample was purified 5 times using three different RNA isolation methods. RNA spike-ins from the RNA Spike-in Kit were added to the lysis buffers of the isolation kits according to the kit handbook. RNA isolation B gives the highest yield, as indicated by the lower Cq values. Overall PCR efficiency is similar between the methods, as determined by the similar UniSp3 results. The high Cq value of UniSp6 from isolation A could indicate the presence of a PCR inhibitor in this sample.

Assessing if samples are affected by hemolysis.
The data from the red blood cell-specific miR-451 and the stable miR-23a can be used to monitor hemolysis. Shown is the difference in the level (ΔCq) of miR-23a and miR-451 for 34 plasma samples. A ΔCq(miR-23a – miR-451) lower than 5 in human serum or plasma represents non-hemolyzed samples. If the ΔCq is close to or higher than 7, there is an increased risk that the samples are affected by hemolysis. In case of high levels of hemolysis, miRNAs from red blood cells will make a significant contribution to the overall miRNA profile identified. Special care must be taken in the analysis of such samples

Panel layout.
The miRCURY LNA miRNA QC PCR Panel contains 6 LNA-enhanced primer sets for quality control of RNA samples (light gray) and PCR efficiency (dark gray). In addition, 5 LNA PCR primer sets that target miRNAs are included. These assays target: miRNAs expressed in most tissues (blue), miRNAs expressed in kidney (purple) and brain (green). Primer sets targeting blood miRNAs are also part of the panel (red). These assays can be used as controls of hemolysis.

The miRCURY LNA miRNA QC PCR Panel offers a unique and comprehensive way of checking sample quality and PCR efficiency. The panel is compatible with many sample types, including cell cultures, tissue samples and biofluids from human, mouse, rat, monkey and dog (see figure miRCURY LNA miRNA QC PCR Panels can be used with a wide range of sample types).

Use the panel to test your isolated RNA to find outliers and suboptimal samples (see figure Determine RNA yields using the RNA Spike-in Kit), including serum/plasma samples affected by hemolysis (see figure Assessing if samples are affected by hemolysis). This saves time and resources in the laboratory.
Principle
The importance of a good start
miRNA qPCR is the most commonly used technique for miRNA expression profiling. There are many advantages of using PCR: it is fast, sensitive and accurate. However, successful qPCR experiments depend on high-quality input RNA. Degraded RNA decreases the efficiency of the technique, and inhibitors can adversely affect the cDNA synthesis and amplification steps. For some samples, such as biofluids, determining the quality of the purified RNA can be difficult.

The miRCURY LNA miRNA QC PCR Panel offers a unique and comprehensive way to check your sample quality and PCR efficiency. The panel contains LNA-enhanced PCR primer sets that target miRNAs and spike-ins that can be specifically used for RNA quality control. These primer sets have been selected based on our extensive experience in miRNA qPCR and analysis of millions of datapoints.

When used in conjunction with the RNA Spike-in Kit, the panel can help you find differences in RNA yield and identify outlier samples (see figure Quickly identify outlier RNA samples). In addition, RNA spike-ins can be used to check the cDNA synthesis and PCR amplification efficiencies.

The miRCURY LNA miRNA QC PCR Panel contains assays for miRNAs that are expressed in a wide range of sample types and is therefore suitable for evaluating the samples in a biological context. Two of the miRNAs in the panel can be used to evaluate the level of hemolysis in samples such as serum and plasma.

Product coverage
The miRCURY LNA miRNA QC PCR Panel consists of 12 predefined assays for use in human, mouse, rat, monkey and dog samples (see figure Panel layout and table miRCURY LNA miRNA QC PCR Panel coverage). The panel can be used for assessing RNA from a wide variety of samples, including tissue, cell culture and biofluids. 

The panel is available in two formats:
  • 96-well, ready-to-use: two 96-well PCR plates containing 16 miRNA QC panels; compatible with most real-time PCR instruments.
  • 384-well, ready-to-use: one 384-well PCR plate containing 32 miRNA QC panels; compatible with most real-time PCR instruments.
The plates are produced in a ready-to-use format, which means that the primer sets are dispensed and lyophilized in the well in an amount sufficient for one 10 µl reaction per well.
