The ALK RGQ RT-PCR Kit is a research use only assay for the detection of ALK kinase domain mRNA expression. Using Scorpions detection technology, the kit provides all necessary reagents optimized for rapid and sensitive detection of low expression of ALK kinase domain mRNA and ABL1 mRNA using real-time RT-PCR on the Rotor-Gene Q 5plex HRM Instrument. The assay uses ABL1 expression as an endogenous internal control.

Principle
The ALK RGQ RT-PCR Kit constitutes a ready-to-use kit for the detection of ALK kinase domain mRNA expression and ABL1 mRNA expression using multiplex, one-step RT-PCR on the Rotor-Gene Q 5Plex HRM instrument. The anaplastic lymphoma kinase (ALK) gene encodes a receptor tyrosine kinase, which belongs to the insulin receptor superfamily. This gene has been found to be rearranged, mutated, or amplified in a series of tumors including anaplastic large cell lymphomas, neuroblastoma, and non-small cell lung cancer. The chromosomal rearrangements are the most common genetic alterations in this gene. The kit enables qualitative measurement of ALK kinase domain mRNA expression and ABL1 mRNA expression from total RNA extracted from formalin-fixed paraffin-embedded (FFPE) samples. Scorpions technologyDetection of amplification is performed using Scorpions. Scorpions are bifunctional molecules containing a PCR primer covalently linked to a probe. The fluorophore in this probe interacts with a quencher, also incorporated into the probe, that reduces fluorescence. When the probe binds to the amplicon during PCR, the fluorophore and quencher become separated. This leads to an increase in fluorescence from the reaction tube.
Procedure
The ALK RGQ RT-PCR Kit comprises a one-step procedure where reverse transcription and PCR are performed in the same reaction tube for both ALK kinase domain mRNA expression detection (FAM probe) and for ABL1 mRNA expression detection (HEX probe). ABL1 is an endogenous control that monitors the quantity and also the quality of RNA used (controls for the presence of inhibitors that may lead to false negative results).
