Investigator Argus X-12 KitFor multiplex amplification of twelve X-chromosomal loci plus Amelogenin
The Investigator Argus X-12 Kit is a unique tool for analyzing X-chromosomal markers, which are highly informative for kinship and paternity testing, and for gonosomal STR analysis of forensic stains. This kit complements the Investigator Argus Y-12 QS Kit, especially in complicated deficiency cases. The genetic analysis is calibrated with BT5 Matrix, an optimized dye technology.
The Investigator Argus X-12 Kit is intended for molecular biology applications in forensic, human identity, and paternity testing. This product is not intended for diagnosis, prevention, or treatment of a disease.
Performance
The Investigator Argus X-12 Kit was validated and evaluated using the GeneAmp 9700 thermal cycler (in standard mode), and the ABI PRISM 310, ABI PRISM 3100, and Applied Biosystems 3130 Genetic Analyzers. The recommended amount of DNA under standard conditions is 0.35 ng. Internal validations demonstrated robust and balanced results with 0.2–0.5 ng DNA, and reliable results with <0.1 ng DNA.
Principle
The Investigator Argus X-12 Kit enables simultaneous amplification of 12 X-chromosomal STR loci (see table "X-chromosomal markers amplified with the Investigator Argus X-12 Kit"), which are highly informative for kinship and paternity testing, as well as population genetics and anthropological studies. The heightened discriminatory power of the kit meets the demands of complicated deficiency cases involving at least one female. In addition, this kit is highly suited for analysis of forensic stains, such as female traces on a male background, and it is an excellent complement to the Investigator Argus Y-12 QS Kit. The markers of the Investigator Argus X-12 are clustered into 4 linkage groups (3 markers per group, see table "X-chromosomal markers amplified with the Investigator Argus X-12 Kit"), and thus each set of 3 markers is handled as a haplotype for genotyping.
Procedure
The 12 polymorphic X-chromosomal loci and Amelogenin are simultaneously amplified in a single PCR and labeled with the dyes listed in the table "Investigator Argus X-12 Kit dyes and corresponding markers". The amplified loci undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as QIAGEN's Investigator IDproof or IDproof Mixture Software.
