Das Investigator IDplex Plus Kit ist ein Multiplex-STR-Assay zur Personenidentifizierung, der die 13 CODIS (Combined DNA Index System) Kernloci, D2S1338, D19S433 und Amelogenin abdeckt.

Das Kit beruht auf unserer Fast-Cycling-Technologie, die eine Amplifikation in etwa 90 Minuten gewährleistet. Das Investigator IDplex Plus Kit ist hochempfindlich und liefert schnelle und zuverlässige Ergebnisse aus DNA-Spuren. Es eignet sich für alle forensischen Anwendungen und Vaterschaftstests. Das genetische Analysegerät wird mit BT5 Matrix kalibriert.

Das Investigator IDplex Kit erfüllt die Anforderungen von ISO 18385.

Performance

Mit Hilfe der Fast-Cycling-Technologie liefert das Investigator IDplex Kit zuverlässige Ergebnisse für STR-Assays in ca. 90 Minuten aus extrahierter DNA.

Der Fast Reaction Mix basiert auf der bewährten QIAGEN-Multiplex-PCR-Technologie, die zur Erhöhung der Inhibitortoleranz optimiert wurde. Die Assay-Leistung, die in Gegenwart steigender Konzentrationen von 6 unterschiedlichen Inhibitoren (Huminsäure, Hämatin, Gerbsäure, Indigokarmin, Kalzium und Kollagen) getestet wurde, zeigt eine hohe Sensitivität und eine überragende Inhibitorresistenz. Dies ermöglicht verlässliche, qualitativ hochwertige Ergebnisse von Fallproben.

Das Investigator IDplex Kit wurde mit dem GeneAmp 9700 Thermocycler und dem Applied Biosystems 3500 Genetic Analyzer validiert und evaluiert.

Die empfohlene DNA-Menge unter Standardbedingungen für die Verwendung mit dem Investigator IDplex Plus Kit beträgt 0,5 ng. Interne Validierungen haben ergeben, dass mit 0,2–2 ng DNA robuste und ausgewogene Ergebnisse und mit < 0,1 ng DNA zuverlässige Ergebnisse erhalten werden.

Principle

Das Investigator IDplex Kit nutzt die Fast-Cycling-PCR-Technologie von QIAGEN für die schnelle, gleichzeitige Amplifikation der 13 CODIS (Combined DNA Index System)-Kernloci, D2S1338, D19S433 und Amelogenin. Sorgfältig entworfene Primer verringern die Überlappung von Loci-Allelen und minimieren so die Wahrscheinlichkeit von Fehlinterpretationen.

Procedure

Die 15 polymorphen autosomalen STR-Marker (D2S1338, D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51, D19S433, D21S11, CSF1PO, FGA, TH01, TPOX und vWA) und das geschlechtsspezifische Amelogenin werden gleichzeitig in einer einzigen PCR amplifiziert und mit einem der in der Tabelle „Farbstoffe und entsprechende Marker des Investigator IDplex Plus Kit“ aufgeführten Farbstoffe markiert.

Yes. The Fast Reaction Mix of the Investigator IDplex Plus Kit contains HotStarTaq Plus enzyme, Mg2+, and dNTPs in a pre-optimized concentration. Only Investigator IDplex Plus Primer Mix and template DNA need to be added. Nuclease-free Water should be added to complete the reaction volume.

Two additional SNP primers have been added to allow balanced amplification of vWA and D16S539 alleles carrying known primer binding site mutations. The mutations covered were observed in concordance studies in samples of African origin at an elevated level. The labelled FGA forward primer has been slightly changed with regard to the linker of the fluorescent dye. The change does not affect PCR amplification, but causes a slight size shift during capillary electrophoresis. The allelic ladder therefore has been changed accordingly.
