Performance

The QIAamp PowerFecal Pro DNA WGS SeqSets and DNeasy PowerSoil Pro WGS SeqSets kits are designed to obtain high-quality DNA, minimize bias and maximize the diversity of the extracted DNA, providing a representative view of the microbiome in the sample. These kits enable precise identification and characterization of the microbial species present in soil or fecal samples.


Unbiased and high-yield isolation of microbial genomic DNA 


DNeasy PowerSoil Pro DNA and QIAamp PowerFecal Pro DNA Kits isolate high yields of DNA from soil and stool/gut samples respectively. 


Both kits identify a greater bacterial diversity compared to other commercially available kits (see figures “Increased bacterial OTUs with the new DNeasy PowerSoil Pro Kit). ” and “Greater total number of bacterial OTUs isolated with the new QIAamp PowerFecal Pro DNA Kit. ”)


Fast and efficient library preparation with low GC bias 


The QIAseq FX DNA Library Kit takes you from 20 pg – 1 μg genomic DNA to sequencer-ready, amplified libraries in just 2.5 hours. It’s faster and easier to automate than mechanical shearing but still generates the high-quality libraries you need for whole genome sequencing of any organism  (see figures “Consistent performance from any input” and “Workflow”).   The workflow reliably generates reproducible DNA fragments of customizable sizes. The QIAseq FX DNA Library Kit performs library preparations with minimal bias by using sequence-independent enzymes for fragmentation – delivering results comparable to mechanical shearing. (see figure“Minimal GC bias compared to other enzymatic methods”)

 
Comprehensive bioinformatic analysis with Microbial Analysis Portal


The Microbial Analysis Portal provides an intuitive, easy-to-use web-based platform allowing researchers that are not bioinformatics experts to analyze the resulting data. You can perform taxonomic identification of microbes, providing a detailed breakdown of the microbial species present in the sample. Additionally, the platform supports AMR analysis, enabling the identification of antibiotic resistance genes and contributing to our understanding of antibiotic resistance spread and evolution. Publication-ready data and figures are easily and quickly generated.

 

Principle

The QIAamp PowerFecal Pro DNA WGS SeqSets and DNeasy PowerSoil Pro WGS SeqSets provide a unified solution for microbiome research. The process begins with the extraction of DNA from the sample using the appropriate kit. The extracted DNA is then subjected to whole genome metagenomic analysis via the QIAseq FX DNA Library Prep Kit, generating a comprehensive dataset that includes all genomic DNA in the sample. This data is then analyzed using Microbial Analysis Portal, which provides detailed insights into the taxonomic composition of the sample and the antibiotic resistance genes present. 

Procedure

Sample extraction


Samples processed with the QIAamp PowerFecal Pro DNA and DNeasy PowerSoil Pro Kits are added to a bead-beating tube. Rapid and thorough homogenization occurs using mechanical and chemical methods. Once cells are lysed, our patent-pending Inhibitor Removal Technology (IRT) is used to remove inhibitors. Total genomic DNA is captured on a silica membrane in a spin-column format. DNA is then washed and eluted, ready for downstream library preparation.


Library preparation


QIAGEN’s QIAseq FX technology incorporates enzymatic DNA fragmentation into a streamlined, optimized protocol that combines fragmentation and adapter ligation in one reaction, saving time and preventing errors. The QIAseq FX kit contains a novel nuclease formulation that digests double-stranded DNA (dsDNA) in a random fashion without sequence preference, generating sequencing libraries with minimal bias. The fragment size can be changed simply by changing the incubation time of the dsDNA with the nuclease, enabling different sequencing applications. Optimized enzyme and buffer compositions ensure high sequencing library yield. Streamlined library construction protocols also enable straightforward automation of library prep on different liquid-handling platforms.


The QIAGEN QIAseq FX DNA Library Kit provides a fast, fully enzymatic procedure, from DNA fragmentation to a next-generation sequencing library, with no cleanup steps until after adapters have been ligated to the sample DNA. 


The QIAseq Normalizer workflow follows a simple bind–wash–elute routine, which is identical for all compatible types of libraries. Normalized libraries are dsDNA and have a common concentration of approximately 4 nmol/L, which allows them to be directly used for pooling and sequencing.

Analysis


After sequencing, samples undergo comprehensive analysis via the Microbial Analysis Portal for taxonomic abundance, differential abundance and AMR-gene abundance.
