Performance
RNAlater Stabilization Reagent可在收集和储存样本时稳定和保护RNA表达谱(参见"RNAlater Reagent prevents degradation of mRNA in tissues")。RNAlater技术对RNA的立即保护可确保下游分析真实地反映整个组织样本的表达谱(参见"RNAlater TissueProtect Tubes prevent RNA changes in tissues")。样本可长期保存,无RNA降解风险,甚至经多次冻融后也无影响。在从多种条件下保存的RNAlater稳定样本中纯化得到的RNA,适用于northern印迹和RT-PCR等下游应用(参见"Stable RNA in tissues: different temperatures and freeze–thaw cycles")。
Principle

一旦生物样本被收集后,其RNA会变得极不稳定。创新的液状RNAlater RNA Stabilization Reagent可快速渗入组织,立即稳定RNA,保护RNA表达模式。还能避免样本处理导致的基因诱导或下调,可进行准确的基因表达分析。

Procedure

组织样本收集后,迅速浸入适当体积的RNAlater RNA Stabilization Reagent中,即可保护RNA,保存样本。用可重复密封的管子,RNAlater TissueProtect Tubes可预先测定所需RNAlater RNA Stabilization Reagent的体积,方便操作和样本保存。该试剂可使RNA在37°C下保存1天,18–25°C下保存7天,2–8°C下保存4周,并且可以在无干冰或液氮条件下处理、储存和运输样本。另外,样本也可以在–20°C或–80°C的条件下存档。

Applications

使用RNAlater RNA Stabilization Reagent稳定的动物和人类组织可用QIAGEN仪器进行样本破碎,并与QIAGEN试剂盒配合使用,进行:
