Performance
可存档的DNA 

DNA的纯度对下游应用结果的准确性至关重要,诸如PCR等敏感的下游应用要求使用最高品质、且分子量尽可能大的DNA,以确保可靠的结果。Gentra Puregene Cell Kit去除污染物和酶抑制剂,纯化获得高度稳定的非常适合存档的DNA。纯化获得的高品质DNA的分子大小为100–200 kb(参见"Reproducible purification of archive-quality DNA")。

Principle
细胞用阴离子洗涤剂裂解,其中加入DNA稳定剂。DNA稳定剂限制胞内DNase的活性,DNA稳定剂也存在于其他环境中。使用RNA降解酶去除RNA。蛋白质等其他污染物通过盐析沉淀法去除。最终,通过乙醇沉淀回收基因组DNA,用水合溶液(1 mM EDTA 、10 mM Tris·Cl pH 7.5)溶解DNA。一般情况下,纯化的DNA的A260/A280 值在1.7–1.9之间,大小可达200 kb。纯化的DNA可贮存于2–8°C、–20°C或–80°C。
Procedure
简单的Puregene流程应用经修饰的盐析沉淀法纯化DNA(参见"Puregene DNA procedure")。无需混合或稀释溶液,并且确保手工操作时间最小化。流程提供方便的实验终止点,可暂时保存部分纯化的样品,安全可靠。
