Performance

Principle

Cignal Reporter Assays含有多拷贝的特定转录因子结合部位与基本的启动子元件,可表达报告基因(萤火虫荧光素酶或GFP)。这些报告基因是功能基因组学和药物研发的有力工具,可检测通路活性。当候选药物、基因敲除、过表达或多肽激活或抑制通路时,荧光素酶或GFP报告基因的活性可被调控,并能被快速、定量检测(参见"Cignal Reporter Assay principle")。Cignal Reporter Assays是利用双报告基因体系的转染即用型载体:双荧光素酶报告基因和GFP报告基因(参见"Cignal Reporter Assay procedure")。

双荧光素酶规格是一种末端式分析,具有卓越的灵敏度、特异性和信噪比。所有的Cignal Reporter Assays都可选这种规格。每次分析都包含通路相关的转录因子报告基因和不可诱导的阴性对照,以及荧光素酶和GFP阳性对照。

GFP规格是监测活细胞通路调控的杰出方法,具有单细胞分辨率。包含信号通路相关的转录因子报告基因和阳性及阴性对照。

Procedure

Cignal Reporter Assays包括预制的、转染即用型通路报告基因(双荧光素酶或GFP),以及阳性和阴性对照。可诱导的通路报告基因和不可诱导的阴性对照都经平行的转染和实验处理。

 双荧光素酶

每个转染子的双荧光素酶结果都计算在内。通过比较处理与未处理转染子报告基因的标准化荧光素酶的活性,确定每个信号通路的活性变化。相同处理的阴性对照作为特异性对照。阳性对照通过检测萤火虫和海肾荧光素酶的表达情况,作为转染效率对照。

GFP

使用流式细胞仪、荧光显微镜或荧光计可对GFP的表达进行定量。通过比较处理与未处理转染子的GFP活性,确定每个信号通路的活性变化。相同处理的阴性对照作为特异性对照。阳性对照通过检测持续表达CMV-GFP报告基因的GFP表达情况,作为转染效率对照。

Applications

Cignal Reporter Assays适用于功能基因组学、蛋白质组学和药物研发,可检测siRNAs、蛋白和小分子化合物对通路抑制或激活的生物效应。
