Principle

QIAamp DNA Mini Kit采用快速离心柱或真空操作,简化了从人类组织样本中纯化DNA的过程。无需苯酚-氯仿抽提。DNA特异性结合到QIAamp硅胶膜上,污染物流走。PCR抑制剂,如:二价阳离子和蛋白,可通过两步有效的洗涤被去除,结合在离心柱上的纯DNA可用水或试剂盒中的缓冲液洗脱。QIAamp DNA技术从人组织样本中纯化得到的基因组、线粒体、细菌、寄生虫或病毒DNA,可用于PCR和印迹实验中。 

Procedure

试剂盒含有优化的缓冲液,能有效裂解样本,可稳定核酸,并促进DNA选择性结合到QIAamp硅胶膜上。将乙醇加入到裂解物中,并上样到QIAamp离心柱上。洗涤缓冲液用于去除杂质,高纯的即用型DNA可用水或低盐缓冲液洗脱。

组织用酶裂解,无需机械匀质化操作,方便的离心柱过程只需20分钟的手工操作时间(裂解时间取决于样品来源)。样品可用离心处理,或已处理过的血液或其他体液可用QIAvac 24 Plus或QIAvac 6S真空底座处理。此外,严密的裂解过程使得QIAamp DNA Mini Kit非常适合于从细菌或寄生物中纯化基因组DNA。在QIAcube全自动核酸纯化仪上纯化基因组DNA用以减少手动操作时间。

真空处理

可用真空装置替代离心机处理血液或体液,可更快更方便地进行DNA纯化。QIAamp Mini离心柱可在QIAvac 24 Plus真空底座上配合VacValves和VacConnectors使用。在样本流速显著不同时,需使用VacValves以确保一致的真空条件。一次性的VacConnectors用于避免交叉污染。使用VacConnectors,也可使QIAamp离心柱操作在带有QIAvac Luer Adapters的QIAvac 6S上进行。 

Applications

QIAamp DNA Mini Kit非常适合于从常规人组织样本中纯化DNA,包括:肌肉、肝脏、心脏、大脑、骨髓和其他组织、拭子(口腔、眼睛、鼻子、咽喉和其他)、CSF、血液、体液和尿液细胞。DNA可在20分钟内从多达25 mg的组织或200 µl的液体中纯化,洗脱体积50–200 µl。
