The BioSprint 96 One-For-All Vet Kit provides automated purification of viral nucleic acids and bacterial DNA from a wide range of veterinary samples on the BioSprint 96. Viral nucleic acids and bacterial DNA are easily and rapidly purified from veterinary samples such as whole blood, serum, plasma, tissue homogenates, cell-free body fluids, and nasal or cloacal swabs. The kit contains all reagents and plasticware necessary for automated nucleic acid purification.

Robust and reproducible purification of viral RNA.
BVDV RNA was isolated from 20 different positive ear notch samples using the BioSprint 96 One-For-All Vet Kit on the BioSprint 96. Viral nucleic acids were amplified in 20 independent experiments using the cador BVDV RT-PCR Reagent on the Applied Biosystems 7500 Real-Time PCR System. BVDV RNA was reliably detected in all experiments, demonstrating the reproducibility of the automated purification procedure.

Sensitive detection of viral RNA in animal blood samples.
Whole blood samples from BVDV positive animals were diluted 10-fold with negative samples as indicated to simulate pooling. Viral RNA was isolated from 50 µl samples using either the BioSprint 96 One-For-All Vet Kit or a kit from Supplier AIV on the BioSprint 96. Purified viral RNA was analyzed using the cador BVDV RT PCR Reagent on the Applied Biosystems 7500 Real-Time PCR System.
Performance
The efficient purification procedure enables robust, reproducible purification of nucleic acids (see figure "Robust and reproducible purification of viral RNA"). Optimized protocols ensure sensitive results in downstream applications. In contrast to a kit from SupplierAIV, viral RNA purified using the BioSprint 96 One-For-All Vet Kit gives lower CT values in real-time RT-PCR applications, enabling more sensitive detection of BVDV (see figure "Sensitive detection of viral RNA in animal blood samples"). With the BioSprint 96 One-For-All Vet Kit, BVDV could be reliably detected in a simulated pool of 100 samples containing just one positive sample.
Principle
The BioSprint 96 One-For-All Vet Kit is used with the BioSprint 96. The kit uses magnetic-particle technology for rapid purification of viral DNA and RNA and bacterial DNA from veterinary samples. The BioSprint 96 controls an array of magnetic rods that can attract or release magnetic particles and transfer them from plate to plate. The sequential transfer of magnetic particles allows a rapid purification procedure to be performed, from the initial binding of DNA to the particles, through to washing of the particles and elution of pure nucleic acids (see figure "Efficient processing of magnetic particles"). Since the instrument transfers magnetic particles instead of liquids, it uses minimal amounts of reagents, enabling cost-efficient sample preparation.
Procedure
Optimized protocols provide reproducible purification of high-quality viral DNA and RNA and bacterial DNA for life science and veterinary pathogen research applications (see figure "BioSprint 96 One-For-All Vet procedure"). Automated processing is fast and, after loading the BioSprint 96, up to 96 samples can be processed in just 24 minutes.

Purified viral DNA and RNA and bacterial DNA are free from PCR inhibitors, enabling sensitive detection in standard, multiplex, and real-time PCR. Purified viral nucleic acids and bacterial DNA are ready to use in downstream assays, including applications in:
