Illumina Whole Genome Expression Profiling Service
The Illumina Whole Genome Expression Profiling Service is intended for molecular biology applications. This product is not intended for the diagnosis, prevention, or treatment of a disease.
Principle

The Illumina Whole Genome Expression Profiling Service offers whole genome gene expression profiling services based on Illumina's industry leading iScan platform and direct hybridization assay-based whole genome expression analysis beadchips. QIAGEN is an Illumina-certified service provider.

Whole-genome expression beadchips allow higher-confidence target discovery, disease classification, and pathway studies. The following Illumina beadchips are used:

HumanHT-12 v4 Expression BeadChip 
MouseWG-6 v2 Expression BeadChip
MouseRef-8 v2 Expression BeadChip 
RatRef-12 Expression BeadChip
Procedure

Recommended sample types and details of sample collection are described below.

For all samples, sample collection should be carried out in a timely manner in an RNase-free environment. Please note the minimum amount of starting materials required (see below for requirements for each sample type). If you have less than the minimum amount or you’ve previously isolated samples that have been stored differently than we recommend, contact QIAGEN Technical Services before submitting your samples.

Before shipping:

Obtain a sample submission form from QIAGEN Technical Services. 
Complete the form and include it with the sample shipment. 
Include the amount of material and its storage conditions. 
Sign and date the form. 
Ship according to the instructions for your sample type
Total RNA

Where total RNA samples are submitted, a minimum of 1 µg RNA per sample is preferred, at a minimum concentration of 50 ng/µl. If samples contain less RNA or the RNA concentration is below 50 ng/µl, please contact QIAGEN Technical Services.

We recommend a Bioanalyzer RIN of no less than 7, if a quality analysis has been performed. QIAGEN will measure RNA concentration and assess the quality of the submitted RNA using an Agilent 2100 Bioanalyzer with an RNA 6000 NanoChip.

If the submitted samples are cells or tissue, Service Core can perform RNA extraction.

Cells

Provide at least 1 million cells for each sample (specify number of cells on the submission form). Snap freeze the cell pellets in liquid nitrogen, store samples frozen at –80°C, and ship on dry ice.

Tissue

Provide at least 50 mg of tissue for each sample (specify the weight of the tissue on the submission form). Snap freeze tissues in liquid nitrogen. Store samples frozen at –80°C, and ship on dry ice.

Sample processing

RNA samples are processed for Illumina gene expression analysis using the following steps:

Sample labeling 
Hybridization
Beadchip wash and staining 
Signal detection 
Beadchip imaging 
Data analysis
Data analysis

The Illumina Whole Genome Expression Profiling Service will provide:

All data related to the quality control and quantitation of the total RNA and amplified RNA samples that were used in the project 
A summary of the internal control features that are part of each Illumina-based gene expression experiment 
The original Illumina iScan output data files (.idat, .jpeg, and .xml files) generated after scanning the BeadChips 
An Excel workbook containing the normalized expression data and lists of 2-fold up- or downregulated genes (generated on GenomeStudio)
In addition, scatter plots, bar plots, line plots, box plots, heat maps and cluster analysis dendrograms are available as custom data analysis options.
