The NeXtal CubicPhase crystallization system enables fully automatable, high-throughput membrane protein crystallization setup using a standard liquid-handling robot or manual setup using a multichannel pipet. The NeXtal CubicPhase Crystallization System includes NeXtal CubicPhase μplates, provided in 1-, 2-, and 3-well formats for increased flexibility in crystallization trial setup. They are available as extra airtight NeXtal Evolution μplates, as 2-well  MRC μplates, and as 3-well IntelliPlate low profile μplates. All plate formats are available precoated with either monoolein or monoolein plus 8% cholesterol. A choice of 2 sets of 96 screening solutions optimized for in meso experiments are also available. NeXtal CubicPhase I Suite contains buffered solutions with different added salts. NeXtal CubicPhase II Suite uses PEGs of differing molecular weight at set pH as precipitating agents.

Workflow for crystallization experiments.
The workflow setup shown is for crystallization on the NeXtal CubicPhase 1w μplate, for the 2w and 3w µplates 100 nl of protein solution is used. The membrane protein solution is dispensed directly onto the dry lipid (monoolein). The protein solution diffuses into and rehydrates the lipid, resulting in the passive formation of the lipidic meso phase.

The NeXtal CubicPhase system.
[A] Protein solution is dispensed onto dried lipids (monoolein). There is an excess of water when the experiment commences. This is known as the sponge phase. [B] Water evaporation triggers concentration of protein and precipitant and also alters the structure of the meso phase. Once an equilibrium in the water pressure at the top of the reservoir and the protein well is reached, the meso phase structure is stabilized in the cubic phase or the lamellar phase. Crystallization will eventually occur if the optimal level of supersaturation is reached and if the protein's three-dimensional organization is correct. Adapted from Caffrey, 2008.

The BR crystal obtained with the NeXtal CubicPhase Cystallization System diffracted at 1.5 Å and allowed confirmation of the structure that was previously solved.

Performance
The innovative NeXtal CubicPhase crystallization system provides an optimized solution for the generation of high-quality membrane protein crystals (see figure "Crystal of bacterio-rhodopsin (BR)"). Only minute amounts of proteins (100 nl) are required compared to standard membrane protein crystallization setup.
Principle
Current methods for the setup of meso phase experiments include numerous manual steps. NeXtal CubicPhase crystallization products enable fully automatable, high-throughput membrane protein crystallization setup using a standard liquid-handling robot. Easy manual setup is also possible using a multichannel pipet. The NeXtal CubicPhase sytem utilizes the advantages of vapor diffusion crystallization together with those of in meso crystallization, which targets the phase transformation point from cubic to lamellar phase, as well as crystallization in the presence of excess water or sponge phase. The NeXtal CubicPhase system uses monoolein (Mo) alone (see figure "NeXtal CubicPhase 1w Mo μplate") or in combination with 8% cholesterol (C) as an additional experiment variable for greater flexibility. Addition of cholesterol allows reduction of the incubation temperature from 22°C to 18°C, which could be a critical experimental factor, especially for the most temperature-sensitive membrane proteins. NeXtal CubicPhase Suites have been tested and approved for compatibility with meso phase crystallization. Dedicated protocols for use with liquid-handling robots and multichannel pipets are included for different plate formats.
Procedure
The NeXtal CubicPhase procedure is easy to perform (see figure "Workflow for crystallization experiments"). It does not require mixing of lipids and the protein solution prior to dispensing.
Applications

NeXtal CubicPhase μplates are compatible with the PX Scanner. The CubicPhase system is highly suited for:

  • Fully automated, high-throughput crystallization
