Product Details

The MinElute 96 UF PCR Purification Kit provides 96-well plates for high-throughput ultrafiltration-based purification of PCR products ≥100 bp in size. The plates are designed to allow elution in very small volumes (as little as 20 μl) delivering high yields of highly concentrated DNA. The procedure can be automated on the BioRobot Universal System. DNA fragments purified with the MinElute system are ready for direct use in many applications, including sequencing, microarray analysis, ligation and transformation, restriction digestion, labeling, microinjection, PCR, and in vitro transcription. The MinElute 96 UF PCR Purification Kit requires the use of the QIAvac Multiwell vacuum manifold (cat. no. 9014579).

The product must be used in conjunction with QIAvac Multiwell.

Performance

The MinElute 96 UF PCR Purification Kit provides high-throughput PCR purification in 96-well plates. Fully automated processing is provided for 1–8 x 96 samples on the BioRobot Universal System or for 1 x 96 samples on the QIAvac Multiwell Manifold (cat. no. 9014579, requires suitable vacuum manifold, e.g., QIAvac Multiwell manifold, plate shaker recommended). The procedure purifies DNA ≥100 bp in size with a recovery of 80–95%. Purified PCR products are eluted directly from the surface of the membrane in small volumes (as little as 20 µl in the manual procedure or 30 µl on BioRobot workstations), providing high-concentration eluates (see figure " Highly concentrated DNA eluates") that are suitable for direct use in a variety of downstream applications. It is possible to process PCR sample volumes up to 150 µl using either a manual or fully automated procedure.

Principle

The QIAGEN MinElute 96 UF PCR Purification Kit combines the convenience of multiwell technology and the separation properties of an ultrafiltration membrane with a unique design. The plates enable the fast and efficient recovery of double-stranded DNA fragments ≥100 bp and the removal of primers (<20mers), unincorporated nucleotides, and salts.

Procedure

MinElute 96 UF PCR Purification Kits provide ultrafiltration technology using vacuum-driven purification on QIAvac manifolds (see flowchart " MinElute 96 UF PCR Purification procedure"). PCR products are loaded into the wells of the ultrafiltration plates, and a vacuum is applied. While small molecules such as primers, salts, and unincorporated nucleotides pass through the membrane, PCR products ≥100 bp are retained. Water, DMSO, or buffers such as SSC can be used for elution. The MinElute 96 UF PCR Purification procedure can also be automated on the BioRobot Universal System.

Can MinElute 96 UF PCR be used to purify PCR samples which contain mineral oil?
We do not recommend the use of samples containing mineral oil since mineral oil will not be removed during the purification procedure.

Can I use a partial MinElute 96 UF plate?

Yes, you may use part of the MinElute 96 UF Plate, and use the rest of the plate later. In contrast to silica membrane based plates, the unused part of the UF plate does not need to be taped off. We do not recommend to re-use MinElute 96 UF Plates more than 2-4 times.
