Investigator Triplex AFS QS Kit

The Investigator Triplex AFS QS Kit is for forensic stain pre-selection, which serves as a starting point for genotyping strategies. The genetic analysis is calibrated with DS-30 matrix.
The Investigator Triplex AFS QS Kit is intended for molecular biology applications in forensic, human identity, and paternity testing. This product is not intended for diagnosis, prevention, or treatment of a disease.
The Investigator Triplex AFS QS Kit was validated and evaluated using the GeneAmp 9700 thermal cycler (in standard mode), and the ABI PRISM 310, ABI PRISM 3100, and Applied Biosystems 3130 Genetic Analyzers.
The recommended amount of DNA under standard conditions is 0.35 ng. Internal validations demonstrated robust and balanced results with 0.1–1.0 ng DNA, and reliable results with <0.1 ng DNA.
The Investigator Triplex AFS QS Kit enables the simultaneous amplification of the two polymorphic STR loci FGA (FIBRA) and SE33 (ACTBP2), and the gender-specific Amelogenin. The kit includes optimized primers and reagents, and the relevant DNA control and allelic ladder.
The QS in the name stands for Quality Sensor. This special feature is an internal PCR control that provides information on the presence of PCR inhibitors and the efficiency of the PCR.
FGA (FIBRA), SE33 (ACTBP2), and Amelogenin are simultaneously amplified in a single PCR and labeled with 6-FAM. The amplified loci then undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as QIAGEN's Investigator IDproof or IDproof Mixture Software. See the table "Technical specifications" for more information.
The Investigator Triplex AFS QS Kit is preferentially used for forensic prescreening strategies or for low-copy and degraded DNA samples that do not give distinct results with complex multiplexes. It enables rapid and reliable DNA genotyping of blood, buccal swabs, and forensic stains.
